Scaffolds of poly-?-caprolacton (PCL) and polyethyleneoxide (PEO) are of big interest in tissue engineering. PEO has the property to enhance the hydrophilicity, by migrating to the surface, and the flexibility of the scaffolds. In this study, the scale-up of the blending process from lab scale to industrial scale, is investigated. In a first stage, blends of PCL and PEO are extruded with a twin screw extruder in the shape of a filament. Because the dimensions of the filament are not constant, no unambiguous conclusions about the properties of the blends can be made. Therefore scaffolds are manufactured with the blends by means of a BioScaffolder. SEM and indentation tests are applied to study respectively the surface and the flexibility of the scaffolds. The conclusion is that the mixing of PEO and PCL is not homogeneous. To enhance the homogenization of the blend, a new method in which the blends are extruded several times, is studied. SEM and SCA shows that the migration of PEO to the surface is not occuring, probably due to the large molecular weight of the used PEO. To know more about the blends, morphologic investigation is done. It is found that the surfaces which are poor in PEO decrease when the blend is extruded more than once. Also the crystals are smaller when the extrusion has been done two times. This means that extruding the blend twice, has a positive influence on the homogenization of the blend. Scaffolds van poly-?-caprolacton (PCL) en polyethyleenoxide (PEO) worden aangewend in tissue-engineering. PEO wordt toegevoegd om de scaffolds meer hydrofiel te maken en de flexibiliteit te verhogen. In dit werk wordt de opschaling van het blending proces van laboschaal naar industriële schaal onderzocht. In een eerste fase worden de blends in een draadvorm geëxtrudeerd met een dubbelschroefextruder. Doordat de afmetingen van de draden niet uniform zijn, kunnen hieruit geen eenduidige conclusies omtrent de eigenschappen van de blends worden getrokken. Daarom wordt overgegaan tot het produceren van scaffolds met behulp van een BioScaffolder. SEM en indrukproeven worden aangewend om respectievelijk het oppervlak en de flexibilteit van de scaffolds te onderzoeken. Hieruit blijkt dat de menging van PCL en PEO in eerste instantie niet homogeen gebeurt. Om de menging te verbeteren wordt vervolgens een nieuwe methode, waarbij de blends meerdere keren geëxtrudeerd worden, ontwikkeld. SEM en SCA tonen aan dat de migratie van PEO naar het oppervlak, wat nodig is om de hydrofiliteit te verhogen, niet gebeurt. De oorzaak hiervan is hoogstwaarschijnlijk het hoge moleculair gewicht van het gebruikte PEO. Morfologie-onderzoek van de blends leert ons dat de PEO-arme zones in de blends kleiner worden naarmate het aantal keren extruderen groter wordt. Tevens worden de kristallen kleiner als de blend twee keer geëxtrudeerd wordt. Dit betekent dat de verdeling van PEO en PCL beter wordt door de blend twee maal te extruderen.
