Abstract

Notre environnement actuel présente de nombreux dangers pour la santé des organismes. L’un d’eux se caractérise par l’altération du système endocrinien des organismes par certaines substances, souvent issues de l’industrie pharmaceutique ou chimique. C’est le cas du bisphénol A. Ce composé chimique, utilisé dans le processus de fabrication de plastiques, se lie aux récepteurs hormonaux et empêchent ceux-ci de fonctionner correctement. En conséquence, l’exposition au bisphénol A augmente la prévalence de l’obésité, de cancers et autres maladies. Aujourd’hui interdit dans certains produits de consommation, le bisphénol A compte de nombreux remplaçants, certainement tout aussi néfaste, comme le bisphénol S. Pour autant, peu de recherches portent leur regard sur le possible effet des bisphénols sur le placenta. Lors du développement embryonnaire, cet organe est le seul moyen de communication avec le monde extérieur, la présence de perturbateurs endocriniens pourrait entraîner des conséquences fortes sur le développement embryonnaire.&#13;Nous avons donc étudié la croissance fœtale et cherché à identifier des marqueurs transcriptionnels placentaires après une exposition au BPA et au BPS à faible dose, chez les rats Wistar. Nous avons alors sélectionné quatre gènes impliqués dans la placentation : Wnt7b, Postn, Grem1 et Spp1 et étudié leur expression dans le placenta. Enfin, nous avons évalué les effets d’une exposition périnatale au bisphénol S sur la croissance postnatale et le développement pubertaire.&#13;Cette expérience nous a permis d’identifier des marqueurs transcriptionnels placentaires après une exposition au bisphénol S, avec une différence sexuelle. L’expression des gènes Spp1 et Wnt7b est diminuées chez les animaux exposés au BPS à faible dose par rapport au groupe contrôle, respectivement chez les femelles et les mâles. Cette altération de l’expression des gènes placentaires est associée à un effet délétère du BPS sur la croissance fœtale qui est significatif chez le mâle. Le BPS diminue également la croissance pondérale postnatale chez la femelle, ce qui semble différer de l’effet obésogène connu du BPA. Concernant la puberté, le bisphénol S semble induire un effet semblable à son prédécesseur puisqu’il induit un léger retard de l’ouverture vaginal chez la femelle.

Abstract

Bisphenol A (BPA) is a synthetic compound for hardening and clearing polycarbonate plastics. BPA is mainly classified as an estrogen-like endocrine-disrupting chemical. In the last decade, attention has arisen in scientific communities that it is not safe to use this chemical in mainly polycarbonate plastics. Exposure to BPA starts in prenatal period, which is the critical period for its toxic effects on different organs. Throughout this book, the readers will obtain information on the effects of BPA on different systems. They will also get information on the prenatal and postnatal effects of BPA. We believe that readers will get qualified scientific knowledge and a general overview of the toxic effects of BPA exposure and its consequences from this book.
