Abstract

The Bunyaviridae is the largest family of RNA viruses, with over 350 isolates worldwide distributed into five genera (i.e., Hantavirus, Orthobunyavirus, Nairovirus, Tospovirus, and Phlebovirus). Many of these viruses are significant human or agricultural pathogens. The increasing number of reports on new emerging bunyaviruses and infection episodes makes it essential that we obtain a comprehensive understanding of bunyaviruses and their infection mechanisms. Although all bunyaviruses possesses a tripartite, negative-sense (or ambi-sense) RNA genome, they exhibit substantial differences in their structure, genome organization and replication strategies, which make functional interpolation across genus boundaries difficult. Fortunately, the bunyavirus field has witnessed many exciting new findings and breakthroughs in recent years. These discoveries span a wide spectrum of research areas, including structural characterization of viruses and viral proteins, the identification of new viruses, investigations into host switch and vectors of transmission, genome-based analysis of virus evolution and phylogenetic lineages, the development of new research tools, such as replicons and reverse genetics, molecular characterization of the virus life cycle at the cell level (i.e., cell entry, replication, transcription, translation, genome packaging, reassortment, and virus assembly, etc.), studies of virus–host interactions and host antiviral defense, the development of vaccines/drugs and the use of bunyaviruses for novel applications. This book includes both research and review papers that together provide a glimpse into the latest research on bunyaviruses and, at the same time, highlight some of the important research achievements made in recent years. Study topics of both a fundamental and applied nature are collated. An informed perspective for future research directions is provided and can stimulate research in some of the understudied areas.

Postgenomic methods, typically based on mass spectrometry, comprise the analysis of metabolites, lipids, and proteins and are an essential complement to genomics and transcriptomics. Multidimensional omics is becoming established to provide accurate and comprehensive state descriptions. This book covers the latest methodological developments for, and applications of integrative multi-omics in biomedical research.

Zika virus (ZIKV), one of the flavivirus family members transmitted by mosquitos, was declared a Public Health Emergency of International Concern by the WHO in February 2016 because of clusters of newborn microcephaly cases and other neurological disorders in Brazil. Most ZIKV infections result in a self-limited flu-like febrile disease, however, if contracted during pregnancy, the virus can also infect fetuses and cause a spectrum of birth defects known as congenital Zika syndrome. To date, no vaccines or antiviral drugs are licensed for ZIKV, and the virus has spread and become endemic to many tropical and sub-tropical countries. Included in this book are thirteen reports addressing diverse aspects of ZIKV–host interactions. These studies range from basic science to clinical research. It is expected that findings from these studies will contribute to a better understanding of the host cells interacting with ZIKV, and may serve as the basis for new diagnostics, antiviral therapies, and vaccine design.

Abstract

Lassa virus causes Lassa fever disease in several countries in West Africa, where it is estimated to infect up to half million people causing roughly five thousand deaths each year. This deadly virus has also been introduced in multiple occasions into the western world, including the United States, United Kingdom, Netherlands, Sweden, and Germany. Lassa virus infection, which is often misdiagnosed, can lead to a wide range of disease symptoms ranging from mild flu-like symptoms to bleeding disorders, multi-organ failure, and death. Despite some major discoveries made in recent years of research on Lassa fever, there are still many unresolved key issues that hamper the development of effective vaccines and therapies. Some of these issues include a detailed understanding of the viral and participating host factors in completing the virus life cycle, in mediating disease pathogenesis or protection, and in activating or suppressing host immune responses against virus infection. This book is devoted to understanding some of these important issues. Expert and timely contributions in the form of editorial and original research and review articles on Lassa fever viral replication, disease pathogenesis and protection, host immune modulations, and other related hot topics are presented in this publication.

Abstract

Alternative treatment modes for antibiotic-resistant bacterial pathogens have become a public health priority. Bacteriophages are bacterial viruses that infect and lyse bacterial cells. Since bacteriophages are frequently bacterial host species-specific and can often also infect antibiotic-resistant bacterial cells, they could represent ideal antimicrobials for fighting the antibiotic resistance crisis. The medical use of bacteriophages has become known as phage therapy. It is widely used in Russia, where phage cocktails are sold in pharmacies as an over-the-counter drug. However, no phage product has been registered for medical purposes outside of the former Soviet Union. The current Special Issue of Viruses contains a collection of papers from opinion leaders in the field who explore hurdles to the introduction of phage therapy in western countries. The articles cover diverse topics ranging from patent to regulatory issues, the targeting of suitable bacterial infections, and the selection and characterization of safe and efficient phage cocktails. Phage resistance is discussed, and gaps in our knowledge of phage–bacterium interactions in the mammalian body are revealed, while other articles explore the use of phages in food production and processing.

Abstract

This compilation of articles elaborates on plant virus diseases that are among the most recent epidemiological concerns. The chapters explore several paradigms in plant virus epidemiology, outbreaks, epidemics, and pandemics paralleling zoonotic viruses and that can be consequential to global food security. There is evidence that the local, regional, national, and global trade of agricultural products has aided the global dispersal of plant virus diseases. Expanding farmlands into pristine natural areas has created opportunities for viruses in native landscapes to invade crops, while the movement of food and food products disseminates viruses, creating epidemics or pandemics. Moreover, plant virus outbreaks not only directly impact food supply, but also incidentally affect human health.
