AIM OF THE STUDY
Mesenchymal stem cells (MSC) are widely used in equine medicine and come from different sources such as bone marrow, adipose and muscular tissue or even cornea. However, when it comes to that source, there is much less literature. The aim of this study is to describe the isolation and culture of corneal stem cells (CSS) as mesenchymal cell source.
ABSTRACT
Regenerative cellular therapies are widely used in equine medicine. Mesenchymal stem cells (MSC) come from different sources such as adipose or muscular tissue and other sources like periodontic ligament, hair follicle or cornea. However, when it comes to that source, there is much less literature. The aim of this study is to describe the isolation and culture of corneal stem cells (CSS).
METHOD
An eye was obtained in necrology service of the Veterinary Medicine Faculty of University of Liege on a horse with unknown date of death and small fragments of limbus were withdraw. Corneal stem cells were isolated and cultured from samples in Dulbecco’s Modified Eagle Medium, Fetal Bovine Serum, glucose and antibiotic.
RESULTS
After one week of incubation at 37°C, a homogenous population of axial, fusiform and fibroblastic-like aspect was observed. Those cells were poorly observed, showing a poor amplification. 
CONCLUSION
Cells isolated from corneal limbus presented a mesenchymal stem cells-like morphology but showed a poor amplification. Different sampling or culture methods may be performed in a future experience to obtain conclusive result. OBJECTIF DU TRAVAIL
Les cellules souches mésenchymateuses (MSC), largement utilisées en médecine équine proviennent de différentes sources telle que la moelle osseuse, le tissu adipeux, musculaire ou encore la cornée. Cependant, lorsqu’on arrive à cette dernière source, la littérature est beaucoup moins étoffée. Le but de cette étude est donc d’étudier l’isolation et la mise en culture des cellules souches cornéennes (CSC) en tant que source de cellules mésenchymateuses. 
RESUME
Les thérapies cellulaires régénératives sont fortement utilisées en médecine équine. On utilise des cellules souches mésenchymateuses (MSC) provenant de différentes sources tel que le tissu adipeux ou musculaire et d’autres sources comme le ligament périodontique, le follicule pileux ou encore la cornée. Cependant, lorsqu’on arrive à cette dernière source, la littérature est beaucoup moins étoffée. Le but de cette étude est donc d’étudier la mise en culture des cellules souches cornéennes (CSC). 
METHODE
Un œil a été prélevé en salle d’autopsie de la Faculté de Médecine Vétérinaire de l’Université de Liège sur un cheval dont la date de mort est inconnue. De petits fragments de limbe cornéen furent prélevés par dissection. Les cellules souches cornéennes ont été isolées à partir des prélèvements et ceux-ci ont été mis en culture dans un milieu contenant du Dulbecco’s Modified Eagle Medium, du sérum bovin fœtal, du glucose et un antibiotique. 
RESULTATS
Après une semaine en incubation à 37°C, une population homogène de cellules présentant un aspect axial, fusiforme et fibroblastique-like, a été observé. Ces cellules étaient faiblement présentes, ce qui montre une faible amplification de celles-ci. 
CONCLUSION
Les cellules isolées depuis le limbe cornéen présentent une morphologie correspondante à celle des cellules souches mésenchymateuses mais ont présenté une faible amplification. Différentes méthodes de prélèvement ou de méthodes de cultures pourraient être mises en place dans une expérience future afin d’obtenir un résultat concluant.

Discussions and debates over the medical use of stem cells and cloning have always had a religious component. But there are many different religious voices.
