Le Syndrome d’Immunodéficience Acquise (SIDA) est provoqué par les virus de l’Immunodéficience Humaine, HIV-1 et HIV-2. Les mécanismes impliqués dans l’évolution de l’infection et la maladie sont encore mal connus. Un travail précédent de notre laboratoire a permis la mise au point d’un modèle in vitro de l’infection par HIV-1 qui pourrait permettre de mieux comprendre les effets du virus sur les cellules infectées. Il a été observé que l’infection par HIV-1 des cellules WE17/10, une lignée de lymphocytes T qui possèdent un phénotype « mémoire », mène, après quelques mois, à un défaut de l’expression du récepteur TCR/CD3 dû à une absence de transcription des ARNm codant pour la chaîne γ de CD3. Suite à ces résultats, on a décidé d’étudier l’effet de l’infection par les virus HIV-2 sur l’expression du TCR/CD3 des cellules WE17/10. Au début, nos expériences ont été retardées par la contamination des cultures par des mycoplasmes.
La lignée WE17/10 infectée par les virus HIV-2ms et HIV-2mvp est productive de virions, mais elle ne l’est pas lorsqu’elle est infectée par le virus HIV-2st. HIV-2 interfère dans l’expression de TCR/CD3. Aux stades initiaux de l’infection, la densité de TCR/CD3 est réduite, donc les cellules infectées TCR/CD3hi+ deviennent TCR/CDlo+ avant de devenir TCR/CD3-. L’absence de la chaîne CD3-γ dans les cellules WE17/10 infectées par HIV-2 conduit à la diminution et la disparition du récepteur TCR/CD3.
L’activation des cellules infectées par HIV-2 avec l’anticorps monoclonal anti-CD3 (OKT.3) montre une perte progressive de réponse cellulaire. Les cellules infectées TCR/CD3hi+ sont encore capables de répondre aux stimulus mais moins que les cellules non infectées. Quand les cellules deviennent TCR/CD3lo+, elles ne sont plus capables de bien répondre à la stimulation avec l’anti-CD3. Les deux phénomènes (TCR-CD3hi+ vers TCR/CD3lo+ et TCR/CD3lo+ vers TCR/CD3-) semblent être le résultat de l’interférence avec la transcription du gène de la chaîne CD3-γ.
Une légère réexpression spontanée de TCR/CD3 a été observée dans les cellules infectées par HIV-2ms ; ceci a été observé occasionnellement dans certaines expériences avec HIV-1. Une réexpression a été obtenue également par activation des cellules infectées au PMA. Ce résultat indique que la perte de l’expression n’est pas la conséquence d’une sélection cellulaire mais peut-être de changements dans des interactions entre produits génétiques viraux et cellulaires.
Il est intéressant de signaler que des études réalisées par d’autres chercheurs sur des lymphocytes de patients asymptomatiques infectés par HIV-1 ont montré des altérations de fonction du TCR/CD3, y compris une perte de réponse à des antigènes de rappel. Il a été montré que ce sont principalement les cellules T mémoire qui ne répondent pas à l’activation de TCR/CD3 et l’interférence du HIV avec cette voie d’activation in vivo est considérée comme un facteur majeur dans la genèse de la perte de réponses immunes cellulaires lors du développement du SIDA
