This medical dissertation explores the neuro-immuno-regulation of inflammation in the colonic mucosa, focusing on mast cells and eosinophils in bowel disorders such as irritable bowel syndrome (IBS) and inflammatory bowel diseases (IBD), including Crohn's disease and ulcerative colitis. The study investigates the role of neuro-immune interactions, particularly involving vasoactive intestinal polypeptide (VIP), in exacerbating mucosal inflammation and disrupting intestinal barriers. Using human and murine colonic tissues, the research identifies increased bacterial translocation in IBS and IBD and highlights the regulatory role of VIP in mast cell activation. The findings aim to enhance understanding of the gut-brain axis and suggest potential biomarkers for treatment outcomes, offering insights into new therapeutic approaches for these chronic bowel conditions.
