In the seventeenth century, smallpox reigned as the world's worst killer. Luck, more than anything else, decided who would live and who would die. That is, until Lady Mary Wortley Montagu, an English aristocrat, moved to Constantinople and noticed the Turkish practice of ""ingrafting"" or inoculation, which, she wrote, made ""the small- pox...entirely harmless."" Convinced by what she witnessed, she allowed her six-year-old son to be ingrafted, and the treatment was a complete success--the young Montagu enjoyed lifelong immunity from smallpox.

Abstract

This book is a compilation of chapters, of which two chapters review the treatment strategies for fibromyalgia syndrome (FMS), and one chapter describes the role of mitochondrial dysfunction and related pathology in the FMS pathogenesis and reviews the appropriate treatment moieties. This book chapter reviews the phenotypic changes that alter the diagnostic criteria and disease activity measures when FMS is comorbid in three potentially painful selected rheumatic diseases. The introductory preface takes the form of an editorial in which I challenge the concept that inflammation, measured as cytokine activity, plays a prominent role in the FMS pathogenesis. All of these chapters and the Preface are authoritative and accomplished discussions that provide novel perspectives on rarely addressed FMS topics.

Autoimmune diseases are diverse and responsible for considerable morbidity. Their etiology remains largely unknown, and current therapy with anti-inflammatory drugs is prone to adverse effects, and rarely curative. New therapies with anti-cytokine antibodies or receptors are promising, but require frequent administration of expensive protein drugs. Gene Therapy of Autoimmune Diseases comprehensively reviews research in gene therapy for autoimmune diseases with viral or non-viral vectors. Gene therapy offers the possibility of long-term, continuous delivery of a wide variety of immunosuppressive, anti-inflammatory, or tolerance-inducing agents. Moreover, highly specific genetically modified cells can be produced. This book discusses the most promising avenues in this exciting new field.
