Abstract

Directed protein evolution-a powerful technique for the discovery of new enzymes and therapeutic proteins-has matured to the point that it is now the centerpiece of enzyme engineering, where it has catalyzed the development of numerous experimental methods. In Directed Evolution Library Creation: Methods and Protocols, seasoned practitioners from many leading laboratories share their best experimental protocols for the generation of molecular diversity. Described in step-by-step detail to ensure experimental success, these protocols include readily reproducible methods for random mutagenesis of entire genes or segments of genes, for homologous and nonhomologous recombination, and for constructing in vivo libraries in bacteria and yeast. In addition to the various protocols for creating libraries, this volume also describes ways to analyze libraries, particularly those made by recombination. An accompanying volume, Directed Enzyme Evolution: Screening and Selection Methods (ISBN: 1-58829-286-X), is devoted entirely to selection and screening methods that can be applied to the directed evolution of enzymes. Taken together, Directed Evolution Library Creation: Methods and Protocols and Directed Enzyme Evolution: Screening and Selection Methods capture for newcomers and more experienced investigators alike all the key methods for using directed protein evolution to better understand protein structure-function relationships, to discover new enzymes and therapeutic proteins, and to design new assays suitable for specific applications.

Abstract

As the use of high-throughput screening expands and creates more interest in the academic community, the need for detailed reference materials becomes ever more pressing. Cell-Based Assays for High-Throughput Screening: Methods and Protocols aims to fill an important part of this need by providing an easily accessible reference volume for cell-based phenotypic screening. Leading researchers in the field contribute state-of-the-art methods with actionable protocols covering four major areas of study: model biological systems, screening modalities and assay systems, detection technologies, and approaches to data analysis. Written in the highly successful Methods in Molecular Biology™ series format, each chapter includes a brief introduction to the subject, lists of necessary materials and reagents, step-by-step laboratory protocols, and a Notes section detailing tips on troubleshooting and avoiding known pitfalls. Cutting-edge and easy-to-use, Cell-Based Assays for High-Throughput Screening: Methods and Protocols presents an overview of relevant approaches, enabling the direct application of existing methods to new discoveries while also inspiring researchers to approach their screening projects in a conceptually modular fashion, enhancing the power to discover through new combinations of existing approaches.

Abstract

Chemical library technologies have brought about dramatic changes in the drug discovery process, and, though still evolving, they have become an integral part of ongoing drug discovery research. In Chemical Library Design, experts in the field provide methods and detailed protocols delving into this key process of selecting useful, biologically relevant compounds from large pools of synthesizable candidates. This compendium includes chapters on historical overviews, state-of-the-art methodologies, including structure-based and fragment-based library design, practical software tools, and successful and important applications of chemical library design. As a volume in the popular Methods in Molecular Biology™ series, the thorough contributions provide the kind of meticulous description and implementation advice that is crucial for getting optimal results. Authoritative and cutting-edge, Chemical Library Design is an ideal reference for all scientists seeking the technology needed to aid in the search for new and vital drugs.
