The expression of the mRNA of the « Endometrial Bleeding Associated Factor » (EBAF), also called TGF-β4 and LEFTY A, is increased in human endometrium at the end of the menstrual cycle when the fall of estrogens and progesterone plasma concentration triggers menstruation. In the present study, the relation between these two events was studied using endometrial explants, cultured with or without 17-β estradiol and progesterone, and in which the amount of EBAF mRNA was quantified by competitive RT-PCR. This high performance technique was chosen because of the small amount of tissues in the biopsies. First, it was adapted to our experimental model and its reliability and reproducibility were investigated. Secondly, using this technique, no clear effects of the ovarian steroids on the expression level of EBAF mRNA were observed in cultured endometria. Moreover, no relation was observed between these results and the expression of other proteins, matrix metalloproteinases (MMPs) and tissues inhibitor of matrix metalloproteinases (TIMPs), which have been shown in our laboratory to be highly regulated by estrogens and progesterone L’expression de l’ARNm de l’”Endometrial Bleeding Associated Factor” (EBAF), également appelé TGF-β4 et LEFTY A, dans l’endomètre humain culmine à la fin du cycle menstruel, au moment où la chute de la concentration plasmatique des œstrogènes et de la progestérone provoque le déclenchement de la menstruation. La relation entre ces deux événements a été étudiée au moyen d’explants d’endomètres, cultivés en présence ou en absence de 17-β œstradiol et de progestérone, et dans lesquels le taux d’ARNm d’EBAF a été quantifié par RT-PCR compétitive. Cette technique performante a été choisie en raison de la taille réduite des échantillons. Dans un premier temps, elle a été adaptée à notre modèle expérimental et sa fiabilité et sa reproductibilité ont été démontrées. Dans un second temps, son application expérimentale n’a pas montré d’effets évidents des stéroïdes ovariens sur le niveau d’expression de l’ARNm d’EBAF dans l’endomètre en culture. D’autre part aucune relation n’a été constatée entre nos observations sur EBAF et l’expression d’autres protéines, les métalloprotéases matricielles (MMPs) et les inhibiteurs tissulaires des métalloprotéases matricielles (TIMPs), dont la régulation par les œstrogènes et la progestérone a largement été démontrée au laboratoire
