La toxicité du cisplatine fait intervenir de nombreuses composantes telles que l’apoptose (rôle important de -53 avec la voie mitochondriale), l’inflammation (rôle important de TNF-α), modulation d’enzymes, production d’espèces oxydantes… Différentes stratégies existent pour diminuer cette toxicité en modifiant l’expression des ces constituants.
