Abstract

The OECD Guidelines for the Testing of Chemicals is a collection of about 150 of the most relevant internationally agreed testing methods used by government, industry and independent laboratories to identify and characterise potential hazards of chemicals. They are a set of tools for professionals, used primarily in regulatory safety testing and subsequent chemical and chemical product notification, chemical registration and in chemical evaluation. They can also be used for the selection and ranking of candidate chemicals during the development of new chemicals and products and in toxicology research. This group of tests covers physical-chemical properties.

Abstract

Cette Ligne directrice décrit un essai in vitro permettant de détecter les effets de produits chimiques sur la stéroïdogenèse, en particulier la production de 17β-œstradiol (E2) et de testostérone (T). La lignée cellulaire H295R de carcinome surrénalien humain, utilisée pour cet essai, exprime les gènes qui codent toutes les enzymes clés pour la stéroïdogenèse. Après une période d’acclimatation de 24 h dans les plaques de culture multi-puits, les cellules sont exposées pendant 48 h à sept concentrations du produit chimique d’essai, au moins en triplicat. Le solvant ainsi qu’un inhibiteur et un inducteur connus de la production des hormones sont employés à une concentration fixe comme témoins négatifs et positifs. À la fin de la période d’exposition, on analyse la viabilité des cellules de chaque puits. Plusieurs méthodes peuvent être utilisées pour mesurer la concentration des hormones dans le milieu, notamment les trousses commerciales de dosage des hormones ou des techniques instrumentales comme les systèmes combinés de chromatographie en phase liquide et spectrométrie de masse. Les données sont exprimées sous la forme d’un facteur multiplicatif de changement par rapport au témoin solvant et d’une concentration minimale avec effet observé. Si l’essai est négatif, la plus forte concentration testée est indiquée sous la dénomination de «concentration sans effet observé».

Abstract

This Test Guideline describes a laboratory test method to assess abiotic hydrolytic transformations of chemicals in aquatic systems at pH values normally found in the environment (pH 4 – 9). This Guideline is designed as a tiered approach; each tier is triggered by the results of the previous tier.Sterile aqueous buffer solutions of different pH values (pH 4, 7 and 9) are treated with the non-labelled or labelled test substance (only one concentration, which should not exceed 0.01 M or half of the saturation concentration). They are incubated in the dark under controlled laboratory conditions (at constant temperatures). After appropriate time intervals, buffer solutions are analysed for the test substance and for hydrolysis products. The preliminary test should be carried out for 5 days at 50 ± 0.5°C and pH 4.0, 7.0 and 9.0. The second tier consists of the hydrolysis of unstable substances, and the third tier is the identification of hydrolysis products. The higher Tier tests should be conducted until 90 % hydrolysis of the test substance is observed or for 30 days whichever comes first.

Abstract

This Test Guideline describes the determining of the ultraviolet-visible (UV-VIS) absorption spectrum of a chemical compound to have some indication of the wavelengths at which the compounds may be susceptible to photochemical degradation. Degradation will depend upon the total energy absorbed in specific wavelength regions. The absence of measurable absorption does not preclude the possibility of photodegradation. This method utilises a double-beam spectrophotometer which records only the absorption differences between the blank (the solvent and all present chemical species other than the test chemical) and test solutions to give the spectrum of the chemical being tested. The test should be carried out at 25°C. The test solutions should be made up in a concentration which will result in at least one absorbance maximum in the range 0.5 to 1.5 units.
