Abstract

This book provides a comprehensive overview of the recent developments achieved in the field of chemo/enzymatic cascades with topics spanning from design (in vitro and in vivo) to kinetic- and process modelling as well as process control. Opportunities and challenges of building multi-step chemo/enzymatic reactions are discussed, whereby the latter are critically assessed in each chapter and methods to ease the implementation are explored. Both, multi-enzymatic cascades and chemo-enzymatic cascades are presented with the motivation of combining the strengths of these two worlds (e.g. selectivity, activity and robustness) not neglecting the obstacles and challenges of such endeavour. Furthermore, the use of non-conventional media for catalytic cascade reactions, recent achievements and potential for future developments in a technical environment are addressed.

Abstract

This Microbiology Monographs volume covers the latest advances in laccase applications in bioremediation and waste valorisation. The first three chapters provide a comprehensive introduction to fungal and bacterial laccases (the two most important enzyme groups from an application viewpoint) and their practical use in bioremediation and lignocellulosic waste valorisation. Subsequent chapters discuss possible combinations of laccases and further potentially collaborating enzymes, and offer in-depth insights into laccase immobilisation for wastewater treatment and environmental biosensor applications of laccases. Lastly, the book addresses the quest for enzymes with improved and better-fitting properties, covering laccase engineering by directed and computational evolution, and novel enzymes from extreme environments. As such, it is a fascinating read for microbiologists in both industry and academia. .

Abstract

Proteases and their involvement in cancer progression have been well addressed and documented; however, the emerging premise presented within this book is that Heparanase is a master regulator of aggressive cancer phenotypes and crosstalk with the tumor microenvironment. This endoglycosidase contributes to tumor-mediated remodeling of the extracellular matrix and cell surfaces, augmenting the bioavailability of pro-tumorigenic and pro-inflammatory growth factors and cytokines that are bound to Heparan sulfate. Compelling evidence ties Heparanase with all steps of tumor progression including tumor initiation, growth, angiogenesis, metastasis, and chemoresistance, supporting the notion that Heparanase is an important contributor to the poor outcome of cancer patients and a validated target for therapy. Unlike Heparanase, heparanase-2, a close homolog of Heparanase, lacks enzymatic activity, inhibits Heparanase, and regulates selected genes that promote normal differentiation and tumor suppression. Written by internationally recognized leaders in Heparanase biology, this volume presents a comprehensive understanding of Heparanase’s multifaceted activities in cancer, inflammation, diabetes and other diseases, as well as its related clinical applications to scientists, clinicians and advanced students in cell biology, tumor biology and oncology.
