Abstract

This book summarizes the related research achievements in Antibody-drug conjugates (ADCs) and their cell metabolism kinetics. The book has three main parts. The first part describes the basic theory of ADCs, including the basic concept and structure of ADCs, and the relationship between the targets of ADCs and their specific functions. The second part mainly introduces the endocytosis and intracellular metabolism of ADCs, including the relationship between endocytosis and ADC activity, the endocytosis and intracellular transport of ADCs, the distribution and metabolism of ADC in vivo. Then it discusses the new formats and research technology of ADCs, including the application of miniaturized antibodies in ADC synthesis, novel carriers for ADC design, the technology and application of site-specific conjugation, and approaches for analyzing the drug: antibody ratio (DAR), the study of pharmacokinetics of ADCs. This book combines the basic theory with the research technology. It can be used as a reference book for students, teachers and researchers of biomedical field.

Abstract

Antibody-drug conjugates (ADCs) stand at the verge of a transformation. Scores of clinical programs have yielded only a few regulatory approvals, but a wave of technological innovation now empowers us to overcome past technical challenges. This volume focuses on the next generation of ADCs and the innovations that will enable them. The book inspires the future by integrating the field’s history with novel strategies and cutting-edge technologies. While the book primarily addresses ADCs for solid tumors, the last chapter explores the emerging interest in using ADCs to treat other diseases. The therapeutic rationale of ADCs is strong: to direct small molecules to the desired site of action (and away from normal tissues) by conjugation to antibodies or other targeting moieties. Abstract

The concept of delivering ‘magic bullets’ to treat diseases was first proposed by Paul Erlich in the early 1900’s.  The realization of this concept for the treatment of cancer occurred in the late 1990’s with the approval of monoclonal antibody therapies.  The use of monoclonal antibodies conjugated (linked) to potent cytotoxic agents (antibody-drug conjugates, ADCs) for specifically delivering cytotoxics to cancer cells was an obvious extension of antibody-based therapy.  ADCs have been under intense investigation for several decades; progress, however, has been limited due to toxicity or lack of improved efficacy compared to unconjugated cytotoxics.  More recently, linker technology and target selection have advanced such that several ADCs and immunotoxins are undergoing clinical testing or are approved for use.  This important volume gives the latest and most comprehensive information on the topic, describing different types of ADCs and immunotoxins for both hematologic and solid malignancies. Finally, the volume highlights the promise that this technology holds for diverse types of human cancer.

Abstract

The concept of using bispecific antibodies for cancer therapy by retargeting immune effector cells was developed more than 25 years ago. However, initial clinical studies were rather disappointing mainly due to low efficacy, severe side effects and the immunogenicity of the bispecific antibodies. A deeper understanding of effector cell biology and especially developments in the field of antibody engineering has led to the generation of new classes of bispecific antibodies capable of circumventing many of these obstacles. Furthermore, new applications were established for bispecific antibodies, such as pre-targeting strategies in radioimmunotherapy and dual targeting approaches in order to improve binding, selectivity and efficacy. In this book, the different means of generating bispecific antibodies are described, with an emphasis on recombinant formats, and information on the various applications of bispecific antibodies, e.g. in cellular cancer immunotherapy, radioimmunotherapy and pretargeting strategies, but also on emerging applications such as dual targeting strategies, i.e., the simultaneous inhibition of two targets (cytokines, receptors, etc.) are provided. This book is intended for a broad readership in the field of antibody engineering, mainly from the pharmaceutical and biotechnology sector, but also including academic researchers working in this field.
