Increasing knowledge of the biological is fundamentally transforming what life itself means and where its boundaries lie. New developments in the biosciences - especially through the molecularisation of life - are (re)shaping healthcare and other aspects of our society. This cutting edge volume studies contemporary bio-objects, or the categories, materialities and processes that are central to the configuring of 'life' today, as they emerge, stabilize and circulate through society. Examining a variety of bio-objects in contexts beyond the laboratory, Bio-Objects: Life in the 21st Century explores new ways of thinking about how novel bio-objects enter contemporary life, analysing the manner in which, among others, the boundaries between human and animal, organic and non-organic, and being 'alive' and the suspension of living, are questioned, destabilised and in some cases re-established. Thematically organised around questions of changing boundaries; the governance and regulation of bio-objects; and changing social, economic and political relations, this book presents rich new case studies from Europe that will be of interest to scholars of science and technology studies, social theory, sociology and law.

The past decade has witnessed a spectacular explosion in both the development and the use of transgenic technologies. In Transgenesis Techniques: Principles and Protocols, Second Edition, authoritative researchers-including members of the Dolly team-take stock of these exciting developments to present a collection of readily reproducible techniques for the generation and analysis of transgenic animals, focusing on the manipulation of the mammalian genome. This second edition devotes much space to the creation of genetically modified murine strains, providing access to the germline both by conventional pronuclear injection and by retroviral and adenoviral infection. Extensive coverage is also given to the generation, maintenance, and manipulation of embryonic stem cell lineages, with protocols for both constitutive and conditional (Cre-lox or the Flp-frt systems) gene targeting. Additional chapters provide cutting-edge techniques for the cryopreservation of both male and female germlines, for the generation of transgenic sheep by nuclear transfer, and for the basic analysis of transgenic organisms. Comprehensive and highly practical, this second edition of Transgenesis Techniques: Principles and Protocols updates and expands the acclaimed first edition with a wealth of easy-to-follow cutting-edge protocols and practical tips for producing transgenic animals and analyzing their integrated transgenes.

Over the past twenty years, the disciplines of cellular and molecular biology have generated a bounty of information pertaining to the understanding of the structure and function of eukaryotic cells . It is now recognized that individual cells regulate their activities in response to a multitude of external signals ; that external signals, a- ing through a limited number of second messenger systems, modulate the cell's pattern of gene expression; and that the cohort of genes available for regulation is dependent upon the developmental history of the cell. Extrapolating these truths to the whole organism is a challenge that has been met by an increasing number of investigators . The wealth of information that molecular and cell biology has provided us with is now being integrated into an understanding of whole t- sues, organs, and organisms. Without such an integration, the molecular information will be nothing more than a confusing catalog of sequences and structures. Central to this project will be the app- cation of the powerful new techniques described in Transgenesis Techniques: Principles and Protocols .

Nuclear Transfer Protocols: Cell Reprogramming and Transgenesis is a comprehensive review of nuclear transfer technology in vertebrates, aimed at reprogramming differentiated nuclei and effecting targeted gene transfer. The authors provide readily reproducible techniques for the generation of cloned embryos and animals in a number of key research and commercially important vertebrates. Additional chapters provide alternative cutting-edge methods for nuclear transfer, such as zona-free nuclear transfer. Of immense benefit are descriptions of procedures associated with cloning, such as in vitro maturation of oocytes, activation and culture of cloned embryos, maintenance of pregnancy, and neonatal care of clones.
