This book provides a comprehensive overview of he basic mechanisms underlying areas of acute leukemia, current advances, and future directions in management of this disease. The first section discusses the classification of acute leukemia, taking into account diagnoses dependent on techniques that are essential, and thankfully readily available, in the laboratory. The second section concerns recent advances in molecular biology, markers, receptors, and signaling molecules responsible for disease progression, diagnostics based on biochips and other molecular genetic analysis. These advances provide clinicians with important understanding and improved decision making towards the most suitable therapy for acute leukemia. Biochemical, structural, and genetic studies may bring a new era of epigenetic based drugs along with additional molecular targets that will form the basis for novel treatment strategies. Later in the book, pediatric acute leukemia is covered, emphasizing that children are not small adults when it comes to drug development. The last section is a collection of chapters about treatment, as chemotherapy-induced toxicity is still a significant clinical concern. The present challenge lies in reducing the frequency and seriousness of adverse effects while maintaining efficacy and avoiding over-treatment of patients.

Better therapy of acute leukemias depends ultimately on better understanding of the distinction between leukemic and normal progenitor cells. Thus, several chapters in this volume describe current knowledge of acute leukemia biology and discuss new classification systems that have arisen consequent to emerging insights into pathogenesis. Treatment has also improved for particular types of acute leukemia. Given our intention that the book be useful to clinicians, we pay particular attention to these advances, to management of acute leukemia in general, and to development of new therapies.

Childhood acute leukemias are one of the main causes of death in children aged 1 to 14 years in some countries; and unfortunately, we have been unable to prevent it. Certainly, a good parcel of it is due to the poor understanding about its etiology. This book aims to describe the most important theories and hypothesis regarding childhood acute leukemia. Written by the most outstanding researchers in the field, this book intends to contribute to a greater understanding of the etiology of this disease. It goes beyond the simple and common analysis of risk factors, which hardly allows us to draw definite conclusions. By addressing the etiology of the disease, discussing from molecular biology until epidemiology and clinical manifestations, this book will guide present and future approaches, contributing for a better clinical management of leukemia in children. The knowledge regarding etiology is a crucial step for a better evaluation, prevention and treatment of a disease.
