Tesamen met de stijgende consumentenvraag voor verse levensmiddelen met een hoge microbiële veiligheid en kwaliteit is in de levensmiddelenindustrie een shift van de meer traditionele, thermische bewaartechnieken naar alternatieve (niet-thermische) technologiëen waar te nemen. Organische zuren zijn in deze context zeer populaire bewaarmiddelen. Ze worden vaak bewust als antimicrobieel additief toegevoegd aan minimaal behandelde levensmiddelen, of kunnen in situ in het levensmiddel geproduceerd worden door fermentatieve of protectieve culluren. Aangezien de antimicrobiële werking van organische zuren voornamelijk bestaat uit een verlaging van de externe pH en een metabolische inhibitie door de ongedissocieerde vorm, is een wiskundige (predictieve) modellering van deze effecten op de microbiële evolutie wenselijk om de microbiële veiligheid van levensmiddelen te kunnen bepalen. In dit onderzoekswerk is de invloed van pH en melkzuur op het microbiële inactivatieproces bestudeerd. Twee gevalstudies zijn hierbij onderzocht. In een eerste wordt melkzuur geproduceerd door een melkzuurbacterie in mengcultuur met een pathogeen organisme. Een model voor de melkzuurgeïnduceerde microbiële inhibitie is uitgebreid naar de daaropvolgende inactivatie van het targetorganisme. Hierbij is de biologische kennis dat zowel pH als ongedissocieerd melkzuur een invloed hebben op de microbiële evolutie in rekening gebracht. In de tweede gevalstudie is melkzuur als antimicrobieel additief aan het experimentele medium toegevoegd. De individuele, ongecorreleerde effecten van de beïnvloedende factoren pH en [LaH] op het microbiële inactivatieproces zijn bestudeerd. Modelstructuren zijn ontwikkeld die de microbiële inactivatie kunnen voorspellen als functie van pH en [LaH]. Together with the increasing consumer's demand for fresh-like food products with a high safety and quality level, the food industry is shifting from the more traditional thermal preservation techniques to alternative (non-thermal) technologies. In this context, organic acids are popular preservatives. They are sometimes deliberately applied in minimally processed foods as an antimicrobial additive, or can be produced in situ by fermentative or protective cultures. Since the organic acid antimicrobial activity is mainly attributed to a lowering of the external pH and metabolic inhibition through its undissociated form, a mathematical (predictive) modeling of these effects on the microbial evolution might be desirable in determining the microbial safety of these food products. In this dissertation, the influence of pH and lactic acid on the microbial inactivation process is investigated. Two case studies are explored. In the first, lactic acid is produced by a lactic acid bacterium in coculture with a pathogen. A model describing the lactic acid induced microbial inhibition is extended to describe the subsequent inactivation of the target organism. Herein, the biological knowledge that both pH and undissociated lactic acid ([LaH]) have an influence on the microbial evolution is incorporated. In the second case study, lactic acid is added as an antimicrobial additive to the experimental medium. The individual, decorrelated effects of its influencing factors pH and [LaH] on the microbial inactivation process are investigated. Model structures are developed to describe the microbial inactivation process as function of pH and [LaH]. Samen met de stijgende consumentenvraag voor meer verse levensmiddelen met een hoge sensorische en nutritionele kwaliteit is in de levensmiddelenindustrie een groeiende interesse ontstaan voor alternatieve behandelings- en bewaartechnieken. Organische zuren zoals melkzuur en azijnzuur zijn in dit opzicht zeer populaire bewaarmiddelen. Ze vertonen een zeer efficiente remmende en/of afdodende werking ten opzichte van pathogenen en bederforganismen en dit reeds bij lage concentraties. Aangezien organische zuren bovendien gekenmerkt worden door een veiligheidsstatus vormen ze een interessant alternatief voor bepaalde chemische en fysische bewaartechnieken. In dit doctoraatsproject wordt de invloed van het organische zuur melkzuur op de microbiële afdoding bestudeerd. Hiertoe worden twee gevalstudies bestudeerd. In een eerste wordt melkzuur geproduceerd door een melkzuurbacterie. Een wiskundig (voorspellend) model dat de melkzuurgeïnduceerde remming van het doelorganisme kan beschrijven is uitgebreid naar een model dat ook de daaropvolgende afdoding van het betreffende microorganisme kan voorspellen. In een tweede gevalstudie wordt melkzuur toegepast als antimicrobieel additief. De afzonderlijke effecten van pH en de ongedissocieerde vorm van het zuur zijn uitvoerig bestudeerd en vertaald naar een wiskundig model. Together with the increasing consumers' demand for more fresh-like food products with a high sensorial and nutritional quality, there is a growing interest in the food industry for alternative food processing and preservation techniques. In this respect, organic acids (e.g., lactic acid, acetic acid) are popular preservatives. They are effective against a broad spectrum of pathogenic or spoilage microorganisms, even at low concentrations. Moreover, since organic acids are characterised by a safe status, they are an interesting alternative for chemical and physical preservation techniques. In this dissertation, the influence of the organic acid lactic acid on the microbial inactivation process is investigated. Two case studies are explored. In the first, lactic acid is produced by a lactic acid bacterium. A mathematical (predictive) model describing the lactic acid induced inhibition of the target organism is extended to describe the subsequent inactivation of the latter organism. In a second case study, lactic acid is applied as an antimicrobial additive. The individual effects of pH and the undissociated form of the acid are extensively investigated and translated into an appropriate predictive model.
