Abstract

Novel molecular motifs named Immunoreceptor Tyrosine-based Inhibition Motifs (ITIMs) have recently been recognized in the intracytoplasmic domains of a still-increasing number of receptors which control cell activation and proliferation. Research on ITIM-bearing molecules has developed exponentially during the last three years, generating new concepts with important consequences in basic research and with exciting potential clinical applications. The present volume contains 15 reviews written by authors who all made significant contributions to the identification of ITIM-bearing molecules and the study of their biological properties. It constitutes the first synthesis ever published that is specifically devoted to this emerging topic.

Abstract

Immunologists today are interested in all of the diverse cell-types involved in host defense and have a deeper appreciation of the importance of innate immune mechanisms as a first line of protection against pathogens. This volume thus discusses the isolation and functional characterization of cells involved in innate immunity in mouse and man, including mast cells and eosinophils. Other focuses include natural killer cells, methods in statistics, in vivo imaging, genome engineering, and mutagenesis and culture that are adapted to the study of innate immunity in these hosts. These are complemented with a series of chapters dealing with alternative models: plants, worms, mosquitoes, flies, and fish. Together, these approaches and models are being used to dissect the complex interplay between hosts and pathogens and contribute to developing strategies to help fight infection. With chapters written by experts on the cutting-edge of this technology, Innate Immunity is an essential reference for immunologists, histologists, geneticists, and molecular biologists.

Abstract

Si son rôle de bouclier est évoqué depuis des années, la réponse immunitaire antitumorale n'a pu être comprise que très récemment. Les armes extraordinairement précises que sont les thérapies ciblées offrent à la recherche de nouveaux angles d'attaque. La mise au point d'anticorps ciblant les points de contrôle du système immunitaire, ainsi que le développement des lymphocytes T exprimant des récepteurs d'antigène " chimériques ", modifient radicalement l'approche tactique. C'est une véritable lame de fond, ouvrant des perspectives inédites dans le contrôle de pathologies tumorales qui apparaissaient sans issue. La compréhension des mécanismes d'action de ces nouveaux outils thérapeutiques est essentielle pour mieux définir leurs indications et imaginer de nouvelles combinaisons, l'enjeu étant d'identifier la meilleure stratégie en fonction de la tumeur et du malade. NK cells have travelled a circuitous path from their initial description as ‘spontaneous killers’ (for some simply an experimental artifact) to being a bona fide subset of innate lymphoid cells with a complementary mode of action in immune defense and an important mediator of immune reactivity in health and disease. Together, these reviews provide a timely and concise picture of the evolution of NK cells as essential agents in immunity and as potent weapons against disease. This book offers an appealing and insightful resource for scientists and clinicians.
