Abstract

The book describes most of the methods that are currently used in metastasis research. Both in vivo and in vitro protocols are illustrated, so that the metastatic process can be either analysed as a whole, or single events addressed separately. Each method is described in the frame of the metastatic process, therefore its significance and its limitations in the context of metastasis are always taken into account. Whenever possible, several alternative procedures are reported per each experimental issue, so that the researcher can choose the one that better suits her/his needs and possibilities. During the past 30 years a big effort has been made to elucidate the molecular mechanisms of cancer metastatis, the leading cause of death for cancer patients. A considerable number of assays have been set up, that can be used to address specific questions concerning the single metastatic steps, or can be applied to develop and test drugs specifically interfering with selected events during the metastatic spread. This book contains an exhaustive description of most of the methods and their rationale, that are currently used in metastatic research, both to analyse metastasis in its entirety (in vivo models), or to dissect the single steps of the metastatic process (in vitro assays). Part II of a two-volume set, dealing with diagnostic methods of nucleic acid hybridization. This work covers probe labelling and hybridization techniques. Immobilized pH gradients (IPG) represent the most advanced development of isoelectric focusing (IEF). Originally developed to overcome all the problems of IEF in soluble amphoteric buffers (CA) (such as pH gradient instability, complexation with CA chemicals, unreproducibility of pH gradients, protein precipitation at the pI), it turned out to be an entirely new technique, quite different in principle and operation from conventional IEF. The book is thus meant to bring the reader up to date with this fast developing field. The book is divided into six chapters containing information on: detail. The central role of RNA in many cellular processes, in biotechnology, and as pharmaceutical agents, has created an interest in experimental methods applied to RNA molecules. This book provides a comprehensive collection of tested manuals for investigating RNA-protein complexes in vitro. Offers a review of developments along with tried and tested methods for isolation, resolution and quantification of inositol phospholipids and inositol polyphosphates in both cells and tissues. This book includes detailed methodology for identification of molecular species of inositol phospholipids, including their phosphates and glycans. Describes the use of synthetic peptides in molecular biology and practical protocols on how to conjugate peptides, immunize animals with peptides and monitor immune responses to peptides in vitro. This book provides information on antigenic specificity, prediction of antigenic sites in proteins and applications of peptides in immunology.
