This dissertation by Hanna Jonasson presents a model-based quantitative assessment of skin blood flow and oxygen saturation using combined diffuse reflectance spectroscopy (DRS) and laser Doppler flowmetry (LDF). The research aims to develop an optical method for accurately measuring oxygen saturation, red blood cell (RBC) tissue fraction, and speed-resolved perfusion in absolute units. The study evaluates different calibration methods and model assumptions in optical phantoms and skin measurements. The integrated system demonstrates potential clinical applications for assessing microvascular function in both healthy individuals and patients with type 2 diabetes, showing reduced RBC tissue fraction and nutritive blood flow in patients compared to controls.
