This thesis is devoted to developing post-Ugi transformations involving gold-catalyzed dearomative ipso-cyclization as a key sequence for the construction of natural or drug-like complex small molecules in two operational steps. The first step is the Ugi-4CR, which can generate diversity and the first stereogenic center, while the second step, the efficient gold triggered dearomative cascade reaction, delivers diverse complex heterocycles with multiple stereogenic centers. Chapter 1 of this thesis will demonstrate the background of related chemistry.Chapter 2 describes a post-Ugi gold(I)-catalyzed dearomatization/ipso-cyclization/aza-Michael addition sequence for the synthesis of complex tetracyclic benzo[e]pyrrolo[2,3-c]indole-2,4,7(5H)-triones from readily available building blocks with unique diastereoselectivity.Chapter 3 describes a post-Ugi gold(I)-catalyzed dearomatization/ipso-cyclization/Michael addition sequence for the construction of diverse (hetero)arene-annulated tricyclic heterocycles containing indole, pyrrole, (benzo)furan, (benzo)thiophene, pyrazole, and electron-rich arene groups.Chapter 4 is dedicated to elaborate a modular access to diverse bridged indole alkaloid mimics involving an efficient cationic gold-triggered intramolecular cascade non-oxidative dearomative spirocarbocyclization/concerted [4+2] cyclization sequence as key strategy, furnishing the architecturally complex and distinct bridged heterocyclic scaffolds with good diastereoselective control.Chapter 5 depicts a gold-catalyzed post-Ugi dearomative spirocyclization/1,6-addition sequence for the rapid synthesis of diverse fused polyheterocyclic scaffolds bearing indole, pyrrole, benzothiophene, furan or electron-rich arene moieties from easily available building blocks.Finally, general conclusions and perspectives are highlighted.
