Given the recent advances in methods for characterizing brain structural and functional connectivity in vivo using MRI, there has been increased interest in leveraging the integration of complementary, multimodal data to better characterize connectivity changes. Specifically, a suitable technique should operate on data that is realistically obtainable in a clinical setting, be data-driven and robust to modeling choices, and provide quantitative results that are easily interpretable by the medical professional. As a first step towards meeting these requirements, our starting point was the recently proposed track-weighted imaging (TWI) contrast mechanism, using functional connectivity (FC) maps as base maps (i.e. TW-FC). A significant limitation of this approach is that it requires the use of a group-level mask to be robust to noise and FC map variability. To eliminate the need for prior knowledge about FC network topology and for finding a hard criterion to separate `irrelevant' FC clusters, we applied threshold-free cluster enhancement (TFCE) to FC maps prior to computation of TWI contrast. It was found that the use of TFCE improves the explorative power of TW-FC, without sacrificing much of its robustness and specificity. However, this improvement comes at the cost of altered contrast properties and a more difficult interpretation of TWI scores, which now include information on topological characteristics of FC clusters. We conclude that the use of TFCE could give TW-FC an increased potential to find relevant group differences and to analyze single-subject data, but that further characterization of its properties is needed.
