Glioblastoma, formerly known as glioblastoma multiforme (GBM), represents the highest grade of astrocytomas. The diagnosis of this particularly malignant intracranial tumor with limited metastatic potential is associated with a low average survival (12 to 15 months). ln this context, temozolomide, an alkylating anticancer agent belonging to the class of imidazotetrazines, has gradually established as the chemotherapeutic agent of reference for the management of those tumors. Its use, initially in combination with radiotherapy initially then afterwards as a monotherapy, offers nowadays interesting prospects for the treatment of glioblastomas, and allows an increase in the average survival (14 to 16 months) and in the two­ year survival. Le glioblastome, anciennement appelé glioblastome multiforme (GBM), désigne le plus haut grade des astrocytomes: Le diagnostic de cette tumeur intracrânienne particulièrement maligne mais dont le potentiel métastatique reste limité, n'est associé qu'à une survie moyenne faible (12 à 15 mois). Face à cette problématique, le témozolomide, un agent alkylant anticancéreux appartenant à la classe des imidazotétrazines, s'est progressivement établi comme étant l'agent chimiothérapeutique de référence dans la prise en charge thérapeutique de ces tumeurs. Son utilisation, en association à la radiothérapie dans un premier temps et en monothérapie par la suite, offre de nos jours des perspectives intéressantes dans le traitement des glioblastomes et permet une amélioration notable de la survie moyenne (14 à 16 mois) et de la survie à deux ans.
