This Test Guideline describes a method to determine the amount of pesticide residues which may be accumulated into rotational crops via soil uptake following realistic agricultural practices. These data may be used to establish crop rotation restrictions, for dietary risk assessment and to determine whether maximum residue limits will be needed in rotational crops. Three representative crops should be tested to determine the uptake of residues: these are root and tuber vegetables; leafy vegetables; and small grains. The test uses three rotational intervals. The rotated crops should be planted after the minimum rotational interval that could be expected as part of agricultural practice: 7-30 days, and 270-365 days for crops rotated the following year. The limited field trials should be conducted at two diverse geographical regions. The pesticide should be applied to primary crop or bare soil by the method specified on the pesticide label or proposed label at the maximum label rate and the maximum number of applications. Residues should be analyzed within 30 days of harvesting (and should be stored frozen prior to analysis).

The purpose of this Test Guideline is to determine the effects of a substance administered with food to birds. Birds are fed a diet containing the test substance at a range of concentrations for a period of five days. Two control groups and one treatment group for each of the, at least, five dietary levels of the test substance should be used. Each group consists of 10 birds. The minimum duration of the test is eight days: five days on the test diet followed by a minimum of three days on normal diet. Suitable facilities for holding birds indoors are necessary. These include mechanisms for temperature, humidity and light control as required, as well as pens of suitable capacity for rearing the birds. Mortalities and signs of toxicity are recorded daily. The following observations should be made during the test: signs of intoxication and other abnormal behaviour; mortality; body weights; food consumption.

The method described in this Test Guideline is based on soil column chromatography in disturbed soil. Two types of experiments are performed to determine (i) the leaching potential of the test substance, and (ii) the leaching potential of transformation products in soils under controlled laboratory conditions. At least duplicate leaching columns are packed with untreated, air-dried and sieved soil (< 2 mm) up to a height of approximately 30 cm. Afterwards they are saturated and equilibrated with an “artificial rain” solution and allowed to drain. Then the surface of each soil column is treated with the test substance (non-volatile in water and soil) and/or with aged residues of the test substance. Artificial rain is applied to the soil columns and the leachate is collected. After the leaching process the soil is removed from the columns and is sectioned into an appropriate number of segments depending on the information required from the study. A reference substance (atrazine or monuron) should be used in the leaching experiments. For each soil segment and leachate fraction, the amounts of test substance, transformation products, non-extractables and, if included, of the reference substance should be given in % of applied initial dose.

Cette Ligne directrice décrit une méthode pour déterminer la quantité de résidus de pesticide qui peuvent être accumulés dans des cultures alternées, à la suite de leur absorption dans le sol, dans le contexte de pratiques agricoles réalistes. Ces données peuvent être employées pour établir des restrictions de rotation de récolte, pour l'évaluation des risques alimentaires et pour déterminer si des limites maximum de résidus seront nécessaires dans les récoltes en rotation. Trois plantes représentatives sont examinées pour déterminer les niveaux d’absorption des résidus : ce sont des légumes-racines et tubercules, légumes à feuilles, et petits grains. L'essai emploie trois intervalles de rotation. Les cultures alternées devraient être plantées après un délai de rotation minimal prévisible dans le cadre de la pratique agricole : 7-30 jours, et 270 à 365 jours pour une rotation des cultures l’année suivante. Les études en conditions limitées sur le terrain sont conduites dans deux régions géographiques différentes. Le pesticide est appliqué sur la culture primaire ou le sol nu par le procédé spécifié sur l’étiquette du pesticide, ou l’étiquette proposée, à la dose maximale et au nombre maximal d’applications indiqués. Des résidus doivent être analysés dans les 30 jours suivant la récolte et doivent être stockés congelés avant l'analyse.
