Mechanisms of Adiponectin Action

Abstract

The adipokine adiponectin is very concentrated in plasma, and decreased levels of adiponectin are associated with pathological conditions such as obesity, diabetes, cardiovascular diseases, and metabolic syndrome. When produced in its full-length form, adiponectin self-associates to generate multimeric complexes. The full-length form of adiponectin can be cleaved by the globular form of elastase that is produced locally, and the resulting biological effects are exerted in a paracrine or autocrine manner. The different forms of adiponectin bind to specific receptors consisting of two G-protein-independent, seven-transmembrane-spanning receptors, called AdipoR1 and AdipoR2, while T-cadherin has been identified as a potential receptor for high molecular weight complexes of adiponectin. Adiponectin exerts a key role in cellular metabolism, regulating glucose levels as well as fatty acid breakdown. However, its biological effects are heterogeneous, involving multiple target tissues. The Special Issue “Mechanisms of Adiponectin Action” highlights the pleiotropic role of this hormone through 3 research articles and 7 reviews. These papers focus on the recent knowledge regarding adiponectin in different target tissues, both in healthy and in diseased conditions.

Abstract

The Special Issue entitled “Pediatric and adolescent nephrology facing the future: diagnostic advances and prognostic biomarkers in everyday practice” contains articles written in the era when COVID-19 had not yet been a major clinical problem in children. Now that we know its multifaceted clinical course, complications concerning the kidneys, and childhood-specific post-COVID pediatric inflammatory multisystem syndrome (PIMS), the value of diagnostic and prognostic biomarkers in the pediatric area should be appreciated, and their importance ought to increase.

Abstract

The demand for computational fluid dynamics (CFD)-based numerical techniques is increasing rapidly with the development of the computing power system. These advanced CFD techniques are applicable to various issues in the industrial engineering fields and especially contribute to the design of fluid machinery and fluid devices, which have very complicated unsteady flow phenomena and physics. In other words, to aid the rapid development of CFD techniques, the performances of fluid machinery and fluid devices with complicated unsteady flows have been enhanced significantly. In addition, many persistently troublesome problems of fluid machinery and fluid devices such as flow instability, rotor–stator interaction, surging, cavitation, vibration, and noise are solved clearly using advanced CFD techniques.This Special Issue on “CFD-Based Research and Applications for Fluid Machinery and Fluid Devices” aims to present recent novel research trends based on advanced CFD techniques for fluid machinery and fluid devices. The following topics, among others, are included in this issue:- CFD techniques and applications in fluid machinery and fluid devices;- Unsteady and transient phenomena in fluid machinery and fluid devices;- Pumps, fans, compressors, hydraulic turbines, pump turbines, valves, etc.

Abstract

“About 25 years ago, Mosmann & Coffman introduced the TH1 / TH2 paradigm of T helper cell differentiation which helped explain many aspects of adaptive immunity from eliminating intracellular versus extracellular pathogens to induction of different types of tissue inflammation.   However, TH1 / TH2 paradigm could not adequately explain development of certain inflammatory responses which provided impetus for the discovery of a new subset of T cells called TH17 cells. After the discovery of differentiation and transcription factors for TH17 cells, it was clear that TH17 cells represent an independent subset of T cells with specific functions in eliminating certain extracellular pathogens, presumably not adequately handled by TH1 or TH2 cells. The major role of TH17 cells has been described in inducing auto-immune tissue inflammation.   The discovery of TH17 cells has expanded the TH1 / TH2 paradigm, and the integration of TH17 cells with TH1 and TH2 effector T cells is beginning to explain the underlying mechanisms of tissue inflammation in a number of infections and auto-immune disease settings.”   - From Chapter One by Vijay K. Abstract

With the recent definition of the Th17 cell subset, a new paradigm in innate and adaptive immunity has been established. This book describes the function of the Th17 cell subset and its prevalent role in many inflammatory and autoimmune diseases. The mechanism by which this T cell subset is generated and maintained shows specific differences between mice and man which are reviewed. One characteristic of Th17 cells is to secrete specific and unique cytokines such as IL-17A, IL-17F and IL-22; their functions are described in detail. These cytokines have acquired a prevalent role in many pathological conditions in animal models and in humans. Targeting these cytokines as well as the generation of Th17 cells may open new ways to treat many autoimmune diseases. The role of Th17 in infectious conditions is also addressed.

The book provides state-of-the-art information on the wide spectrum of ways in which cells can respond to different forms of stress induced by chemicals, oxidants, and DNA-damaging agents. Mechanisms are described that involve altered uptake and efflux of chemical agents, intracellular detoxification, and DNA damage responses. Many of these changes trigger a cascade of reactions mediated by stress-activated signaling pathways, which have the capacity to determine whether a cell will survive or die. Abstract

Many research teams are working to demonstrate that microorganisms can be our daily partners, due to the great diversity of biochemical transformations and molecules they are able to produce. This Special Issue highlights several facets of the production of microbial metabolites of interest. From the discovery of new strains or new bioactive molecules issued from novel environments, to the increase in their synthesis by traditional or innovative methods, different levels of biotechnological processes are addressed. Combining the new dimensions of "Omics" sciences, such as genomics, transcriptomics or metabolomics, microbial biotechnologies are opening up incredible opportunities for discovering and improving microorganisms and their production.
