Abstract

"A concise introduction to fundamental principles, this book presents the most important aspects of fluorescence spectroscopy, particularly as they are applied to biology, biochemistry, and biophysics. The author focuses on the essential aspects of fluorescence to encourage readers to design experiments, begin their own fluorescence measurements, and access the literature with greater comprehension. After a general definition of the field and an overview of the nature of light and absorption by biomolecules, the text covers instrumentation, emission spectra, excitation spectra, polarization, anisotropy, quenching, time-resolved methods, and FRET"--

Abstract

Fluorescence imaging is widely used in scientific fields ranging from biology to biomedicine and even materials science. The development of novel fluorescent labels and microscopy techniques has rendered fluorescence imaging profoundly useful. Particularly in bioscience, fluorescence imaging empowers the study of the intracellular distribution, dynamics, gene expression, protein-protein interactions, and protein localization and enables the identification and tracking of lysosomes. Fluorescence imaging is applicable in cells and tissues and is constantly gaining attention in medicine too, in the fields of fluorescence-guided surgery and robotic-assisted fluorescence surgery. Acknowledging all these important new trends, this book provides an overview of the recent advances and applications in fluorescence imaging.

Abstract

This book discusses the use of fluorescence spectroscopy for the quantitative demarcation of glioblastoma, a highly malignant brain tumor, during surgical procedures. The study evaluates the effectiveness of using five-aminolevulinic acid (5-ALA) as a fluorescence contrast agent to differentiate between tumor and healthy brain tissue. The research involves a clinical evaluation at Linköping University Hospital, using a developed fiber-optical fluorescence spectroscopy system. The main challenges addressed include ambient light interference, blood absorption, and the quantification of fluorescence spectra. The findings suggest that fluorescence ratios can indicate tissue type and tumor malignancy degree, potentially improving surgical outcomes and patient survival.

Abstract

"Presenting a detailed, hands-on approach to fluorescence spectroscopy, this book describes experiments that cover basic spectroscopy and advanced aspects of fluorescence spectroscopy. It emphasizes practical guidance, providing background on fundamental concepts as well as guidance on how to handle artifacts, avoid common errors, and interpret data. Nearly 150 experiments from biophysics, biochemistry, and the biomedical sciences demonstrate how methods are applied in practical applications. The result is a hands-on guide to the most important aspects of fluorescence spectroscopy, from steady-state fluorescence to advanced time-resolved fluorescence. Provides a complete overview of nearly 150 experiments using fluorescence spectroscopy, from basic to advanced applications Presents laboratory methods using a variety of instrumental setups with detailed discussion of data analysis and interpretations Covers steady-state phenomena, time-resolved phenomena, and advanced methods Spans biophysical, biochemical, and biomedical applications Describes related concepts, theory, and mathematical background as well as commercially available instruments used for measurements"--
