Abstract

"Through recent research, there has been a significant number of findings involving the biology of macrophages. Beyond the 1st line of defense, macrophages have several specific characteristics across different tissues, organs, and systems. Macrophages comprehend a heterogeneous phagocytic population with wide range phenotypes, and tissue-specific functions, such as bone resorption, extracellular matrix production, red blood cells and bacteria removal, among others. Through innate immune receptors macrophages can modulate several pathways that culminate in the production of various types of inflammatory mediators such as chemokines, cytokines, oxygen reactive species, and lipid mediators which ultimately altered the behavior of other cells. Moreover, environment signals modify and induce metabolic reprograming that it alters phenotype and functions."--

Abstract

De nos jours, l’obésité est devenue un danger majeur pour la santé publique. Favorisant le diabète de type 2, les maladies cardiovasculaires et certains cancers, l’obésité devient une des causes principales de décès prématuré. L’obésité est accompagnée de dyslipidémie, caractérisée par une augmentation des concentrations sanguines en acides gras libres. Cet environnement riche en lipides cause des perturbations immuno-métaboliques sur les cellules sanguines telles que les cellules Natural Killer (NK). Sachant que les cellules iNKT peuvent être activées par la présentation d’antigènes lipidiques endogènes présentés via le CD1d par des CPAs, il est tentant de spéculer que cet environnement pourrait être propice au chargement sur le CD1d des monocytes de lipides antigéniques nouvellement synthétisés ou endocytés. Dans ce mémoire, nous avons donc étudié un éventuel effet indirect des acides gras sur l’activation des cellules iNKT en réalisant des co-cultures de cellules iNKT avec des macrophages THP1-CD1d traités par différents acides gras libres. Nos résultats ont montré qu’il y a bien une activation des cellules iNKT quand elles sont mises en culture avec les macrophages pré-traités spécifiquement avec des acides gras saturés et non avec des acides gras mono-insaturés. De plus, cette capacité des macrophages d’activer les cellules iNKT après un traitement avec les acides gras saturés semble être dépendante de la présentation d’antigènes lipidiques via le CD1d.
