Abstract

Protein phosphorylation via protein kinases is an inevitable process that alters physiological and pathological functions of the cells. Thus, protein kinases play key roles in the regulation of cell life or death decisions. Protein kinases are frequently a driving factor in a variety of human diseases including aging and cellular senescence, immune system and endothelial dysfunctions, cancers, insulin resistance, cholestasis and neurodegenerative diseases, as well as bacterial resistance in persistent infections. Recent developments in quantitative proteomics provide important opinions on kinase inhibitor selectivity and their modes of action in the biological context. Protein Kinase-mediated Decisions Between Life and Death aims to have the reader catch insights about up-to-date opinions on “Protein Kinases” related pathways that threaten human health and life. As “Protein Kinases” are related to many health problems, clinicians, basic science researchers and students need this information. Abstract

Apomixis is the consequence of a concerted mechanism that harnesses the sexual machinery and coordinates developmental steps in the ovule to produce an asexual (clonal) seed. Altered sexual developments involve widely characterized functional and anatomical changes in meiosis, gametogenesis, and embryo and endosperm formation. The ovules of apomictic plants skip meiosis and form unreduced female gametophytes whose egg cells develop into a parthenogenetic embryo, and the central cells may or may not fuse to a sperm to develop the seed endosperm. Thus, functional apomixis involves at least three components, apomeiosis, parthenogenesis, and endosperm development, modified from sexual reproduction that must be coordinated at the molecular level to progress through the developmental steps and form a clonal seed. Despite recent progress uncovering specific genes related to apomixis-like phenotypes and the formation of clonal seeds, the molecular basis and regulatorynetwork of apomixis is still unknown. This is a central problem underlying the current limitations of apomixis breeding. This book collates twelve publications addressing different topics around the molecular basis of apomixis, illustrating recent discoveries and advances toward understanding the genetic regulation of the trait, discussing the possible origins of apomixis and the remaining challenges for its commercial deployment in plants.

Abstract

p38 Mitogen activated protein kinases (p38MAPK) are a group of evolutionary conserved protein kinases which are central for cell adaptation to environmental changes as well as for immune response, inflammation, tissue regeneration and tumour formation. The interest in this group of protein kinases has grown continually since their discovery. Recent studies using new genetic and pharmacological tools are providing helpful information on the function of these stress-activated protein kinases and show that they have an acute impact on the development of prevalent diseases related to inflammation, diabetes, neurodegeneration, and cancer. In this Special Issue we present novel advances and review the knowledge on the identification of p38MAPK substrates, functions, and regulation; mechanisms underlying the role of p38MAPK in malignant transformation and other pathologies; and therapeutic opportunities associated with regulation of p38MAPK activity.

Abstract

In this Special Issue, we have published papers on the health-promoting effects of nutraceuticals from different sources, and their effects in different pathologies. Extracts from plants have been analyzed, for example, extracts from olive leaves, Mikania micrantha, the devil’s claw, raspberries and others, alongside marine phytoplankton, egg-yolk and marketed dietary supplements. The effects of these extracts and dietary supplements have been studied in diseases associated with obesity, and in diseases where inflammation pathways are involved. The effectiveness of resveratrol and curcumin to support the anticancer activity of cisplatin has also been reported, as well as the ability of devil’s claw root extract to stimulate the CB2 receptors in synoviocytes in osteoarthritis patients. The anti-oxidant effect of marine phytoplankton has been studied on muscle damage, both in humans and in an animal model, and the effects of the metabolite of antocianin were analyzed in a mouse model of amyotrophic lateral sclerosis. Finally, reviews on the use of lactoferrin, ω3 and ω6 and abscisic acid have been reported, in addition to the crosstalk between prostate cancer and microbiota inflammation. Although it is not yet possible to draw definitive conclusions on the use of nutraceuticals, several mechanisms of action for many of them have been further clarified.
