Abstract

La cytométrie en flux, technique permettant l'analyse rapide des cellules, a considérablement évolué ces dernières décennies et a contribué à de nombreuses avancées scientifiques et médicales. Ses avancées ont ouvert la porte à des applications devenues incontournables dans différentes spécialités telles que l'hématologie, l'immunologie, la microbiologie, la biologie végétale et marine. La transversalité interdisciplinaire de la cytométrie en flux en fait toute la richesse. La deuxième édition de "La cytométrie en flux" fait le point, en 29 chapitres, sur toutes les nouveautés dues aux avancées instrumentales, à la miniaturisation des lasers, aux progrès de l'optique, de la fluidique mais aussi de l'informatique, des logiciels embarqués et de l'analyse de données, sans oublier les réactifs couvrant maintenant une gamme inimaginable il y a seulement encore quelques années. The cooperation of highly specialized cell types maintains the homeostasis of multicellular organisms. The disturbance of that harmony contributes to the development of several diseases. Most of the cellular functions are executed by proteins, so it is essential to investigate biological processes at the protein level. Antibodies, complex biomolecules with high specificity, are used to recognize our protein of interest in a process known as “immunophenotyping”. One of the routinely used methods to study cellular proteins is flow cytometry, which detects cell surface or intracellular proteins at single-cell resolution. The other most frequent technique is the traditional immunohistochemical investigation of microscopic sections of human tissues. We called authors to publish their latest data studying cancer or autoimmune diseases by immunophenotyping.

Abstract

In many parts of the world, freshwater is a subject of frequent and intense large-scale disturbances. Pollution, water withdrawal, alteration of freshwater flows, road construction, aquifer mining, surface water diversion, desertification, wetland drainage, soil erosion in agriculture, deforestation, and dam building have led to some irreversible species losses and severe changes in community compositions of freshwater ecosystems. Pollution represents one of the most relevant impacts on freshwater environments, ranging from surface water bodies—such as springs, streams, rivers, lakes, and intermittent waterbodies—to groundwater and transitional habitats between surface waters and groundwaters. The origins and fates of pollutants are different and depend on various pollutants, including fertilizers with pesticides in agricultural areas, heavy metals, chlorinated organic compounds, and polycyclic aromatic hydrocarbons (PAHs), which are predominantly derived from industrial and urban settlements. Another pollutant is microplastics, which can increase in concentration in freshwater bodies and constitute emerging contaminants in freshwater systems when taken together with pharmaceuticals, personal care products (PCPs), and endocrine-disrupting compounds (EDCs). The broad distribution of several pollutants leads to significant changes of freshwater ecosystems, together with the extinction of the most sensitive species or the drastic lowering in abundances of others, thus altering community compositions and the ecosystem services provided by freshwater biodiversity.

Abstract

The analysis of circulating tumor cells (CTCs) as a real-time liquid biopsy approach can be used to obtain new insights into metastasis biology, and as companion diagnostics to improve the stratification of therapies and to obtain insights into the therapy-induced selection of cancer cells. In this book, we will cover all the different facets of CTCs to assemble a huge corpus of knowledge on cancer dissemination: technologies for their enrichment, detection, and characterization; their analysis at the single-cell level; their journey as CTC microemboli; their clinical relevance; their biology with the epithelial-to-mesenchymal transition (EMT); their stem-cell properties; their potential to initiate metastasis at distant sites; their ex vivo expansion; and their escape from the immune system.

Abstract

Cyanobacterial abundance has increased disproportionately, and this trend is likely to continue in the coming decades. This increase not only has deleterious effects on ecosystem biodiversity but also adversely affects drinking water supplies, livestock watering, crop yields, aquaculture, etc. Thus, the proliferation of cyanobacterial blooms presents human and animal health risks due to the common production of potent toxins, cyanotoxins. Moreover, these risks are aggravated by the accumulation potential of cyanotoxins and their transference to the food chain. In spite of the worldwide increasing occurrence of cyanotoxins, they are still underestimated in regulations. However, risk management of cyanotoxins is only possible after a thorough risk evaluation, and for that purpose, toxicity and exposure data are required. Thus, occurrence and monitoring information is of key importance, and new data in relation to the conditions that favor cyanobacterial growth and cyanotoxin production are welcome in order to prevent their appearance. On the other hand, in regard to toxicity, there are still many data gaps to fill. This book compiles 10 research papers and a review, which provide valuable contributions on all these aspects and demonstrate the importance of cyanobacteria toxins research.
