Abstract

In diesem Werk werden die richtungsweisenden methodischen Überlegungen des Integrierten Management-Ansatzes für die Umsetzung einer nachhaltigen Unternehmensführung nutzbar gemacht. Dabei wird die Thematik sowohl theoretisch-konzeptionell als auch praxisnah-fallbezogen mit einem intendierten mittelbaren „Brückenschlag“ zwischen beiden Perspektiven ausgeleuchtet. In Teilen der einschlägigen Fachliteratur wird die nachhaltige Unternehmensführung als integrales bzw. integriertes Konzept gekennzeichnet. Der Integrationsfokus sollte sich dabei auf die unterschiedlichen betrieblichen Ebenen und Subsysteme ausrichten. Dem weithin anerkannten St. Galler-Konzept "Integriertes Management" folgend, geht es darum, dass der auf der normativen Ebene postulierte Nachhaltigkeitsgedanke über die Strategie in operative Handlungsvollzüge einfließt und dadurch operationalisiert und für alle Anspruchsgruppen (insbesondere Kunden und Mitarbeiter) konkret erfahrbar wird. Das Konzept „Integriertes Management“ bildet mit seinem metapherhaften Grundverständnis als „Leerstellengerüst für Sinnvolles und Ganzheitliches“ hierzu einen sinnvoll „befüllbaren Rahmen“. Die bewusste Zusammenführung mit diesem Konzept weitet das Integrationsverständnis der Nachhaltigkeit über die konzeptimmanente Verzahnung der konstitutiven Kerntriade (Ökonomie/Ökologie/Soziales) hinaus in alle betrieblichen Subsysteme (Ebenen, Teilfunktionen etc.) hinein. Dieser Herausgeberband mit ausgewählten Autorinnen und Autoren aus Forschung und Praxis ermöglicht darüber hinaus aufgrund der unterschiedlichen Blickwinkel eine intendierte breitgefächerte Ausleuchtung der Thematik mit zum Teil unterschiedlichen Sichtweisen und Standpunkten. Die Herausgeber Prof. Dr. Kristin Butzer-Strothmann ist seit 2011 Professorin für Allgemeine Betriebswirtschaftslehre, insbesondere Marketing und Empirische Sozialforschung an der Leibniz-Fachhochschule in Hannover. Prof. Dr. Friedel Ahlers ist seit 2011 Professor für ABWL mit Schwerpunkten Unternehmensführung und Personalwirtschaft an der Leibniz-Fachhochschule in Hannover.

Abstract

In this study we searched for the common ground between blockchain technology and the cooperative business model. Cooperative firms act up to a certain set of principles and values. One of the reasons for blockchain’s rise in the data storage world seems to be its underlying values. These guaranteed “moral” values make blockchain stand out against centralized data collecting entities. A lot of research has been done concerning blockchain or cooperatives separately, but research comparing both sets of values does not exist yet. Our qualitative research consists of a thorough literature analysis combined with interviews deducted from companies who are building blockchain based applications. Combining both, provided a sufficient foundation to form three linkable value clusters being: Economic democracy, transparency, solidarity. Subsequently, we found one cluster of non-linkable values containing the values of freedom, autonomy/independence and anonymity. The four clusters eventually conclude in some future research questions, which are then bundled in a research agenda. The general conclusion of this study is that blockchain technology can be programmed how one wants it to be and consequently can carry values that one wishes to obtain.

Abstract

Acute myeloid leukemia (AML) is a genetically heterogeneous disease presenting with a wide spectrum of cytogenetic and molecular abnormalities and outcomes. In the first part of this study, we review some main features of fusion gene detection by conventional techniques such as chromosome banding, FISH and RT-PCR. As the number of WHO hematological neoplasms defined by genetic abnormalities and the number of specific treatment approaches that directly or indirectly target specific abnormalities is steadily increasing, there is growing need to simultaneously interrogate a large number of gene targets within a clinically-relevant timeframe. This has directed a shift towards next-generation sequencing (NGS). We discuss several NGS-based approaches for fusion gene detection, focusing on library preparation techniques and data analysis. In the second part of this study we evaluate the current strategy for fusion gene detection in the AZ Sint-Jan Brugge-Oostende tertiary-care hospital and provide some perspectives on the implementation of an NGS-based panel. The primary objective was to determine the frequency of fusion genes detected over a 5-year period using a commercial multiplex RT-qPCR assay for qualitative screening of 28 fusion genes (HemaVision®28Q), and to evaluate concordance between RT-qPCR and chromosome banding/FISH. Based on these results, we estimated if a smaller – and thus less expensive – RT-qPCR panel targeting seven fusion genes (HemaVision®7Q) could be sufficient in our setting. A secondary objective consisted of a proof of principle study for detection of fusion genes using RNA-seq (QIAseq® RNAscan Human Leukemia Panel). In a 5-year period, we detected 28 fusion genes in 225 unique samples from adult patients diagnosed with AML or relapsed AML. We found that the HemaVision®28Q assay performed well as it did not miss any of the included fusion genes when compared to chromosome banding and FISH. In addition, we detected a BCR-ABL fusion with RT-qPCR in one patient sample that was not picked up by chromosome banding or FISH. The HemaVision®7Q would miss 11/28 (39%) of recurrent gene fusions in this 5-year period at our institution, which is not acceptable in our opinion because of the clinical importance of the translocations which are missed. Using four samples we successfully tested an RNA-seq panel, the QIAseq® RNAscan Human Leukemia Panel as proof of principle for NGS-based fusion gene detection to screen more comprehensively for fusion genes in AML using two bioinformatic pipelines. The hands-on-time (three working days) was the main limitation of this technique. Therefore, we see more potential in combined DNA and RNA sequencing. Other possible configurations are broad screening with NGS combined with a smaller RT-qPCR assay such as the HemaVision®7Q or FISH for rapid detection of urgent translocations (e.g. PML-RARA), or automatization strategies for reduction of hands-on-time. Lastly, methods with a short library preparation, such as third generation sequencing, are extremely attractive. Taken together, our study shows that RNA-seq methods are being introduced in clinical diagnostic laboratories for detection of fusion genes and are likely to gain in importance. When partially automated and applied to a larger set of patients it could prove to be a cost-effective method.
