L’essor de l’industrie biopharmaceutique a conduit à un besoin croissant de production de grandes quantités de protéines recombinantes. Une solution pour pallier à cette demande consiste à produire ces protéines d’intérêt sous forme de corps d’inclusion, des agrégats insolubles apparaissant suite à la surexpression des protéines dans les bactéries. Cependant, les corps d’inclusion doivent être solubilisés pour être utilisés, ce qui entraîne la dénaturation de la protéine recombinante. L’étape de renaturation qui s’ensuit est souvent l’étape limitante dans l’utilisation des corps d’inclusion comme moyen de production car aucune technique universelle n’a été découverte à ce jour. Ainsi, le repliement doit être étudié au cas par cas avec chaque protéine. De plus, les rendements obtenus pour des protéines complexes de grandes tailles sont généralement faibles (inférieurs à 30%).
Ce travail s’est focalisé sur le repliement d’une protéine tétramérique complexe produite à partir de corps d’inclusion, la β-lactamase L1, en utilisant la méthode de renaturation en présence du couple SDS-MPD. La détermination des conditions optimales de repliement a été réalisée grâce à un programme de plan d’expérience (Design of Experiment), couplé à l’assistance d’une plateforme de pipetage automatique disponible à Robotein® (http://www.robotein.ulg.ac.be/).
Grâce à cette technique, les différentes conditions physico-chimiques influençant le repliement ont pu être déterminées. Ce travail a également permis de mettre en évidence deux facteurs possédant une importance cruciale lors du repliement de la β-lactamase : l’intégrité structurale des monomères et la pureté de l’échantillon de corps d’inclusion. La présence d’une forme anormale de L1 dans une première production a conduit à une chute drastique du taux de repliement de la protéine. En outre, des corps d’inclusion bruts n’ayant pas subi d’étape de purification ont également conduit à une diminution du taux de repliement de L1. En tenant compte de tous ces paramètres, des taux de repliement de la β-lactamase supérieurs à 70% ont pu être obtenus. Ces résultats de rendements exceptionnellement élevés pour une protéine complexe multimérique confortent le rôle du couple SDS-MPD dans la renaturation de protéines.
