Abstract

The aim of this study was to develop and validate an HPLC-MS/MS assay to determine total and unbound concentrations of temocillin in biological samples. The validated method was used to determine temocillin levels in serum and tissue samples from treated patients.Design and mcthods: Methanolic protein precipitation and ultrafiltration were used for total and unbound concentration extraction. respectively. Extract was injected into a LC-MS/MS system for the quantification of the antibiotic. using ticarcillin as an internal standard.Results: Temocillin total and unbound concentration quantification assays Vere linear over concentrations ranging from 1 to 500mg/L and from 0.5 to 300mg/L respectively. Both assays presented acceptable intra and inter-assay precision and accuracy < 13.9%. Limits of quantification and detection were of l and 0.1Omg/L and 0.5 and 0.05mg/ L for total and unbound concentration respectively. Total temocillin concentration recovery ranged from 85.80 to 99.40%. No memory effect had been observed and temocillin ion suppression effect was <36.2 % in both assays. This method allowed us to determine free and total concentrations in scrum samples from an hemodialysis patient with results similar to those obtained with a previously validated HPLC-UV method for total concentration. lt was then applied to serum and pancreatic samples from an intensive care patient in whom we observed a high free fraction (related to hypoalbuminemia) and a tissular concentration higher than scrum levels.Conclusion: The method described is fast, sensitive and selective. with no interferences. This method may be used for bot h pharmacokinetic studies and therapeutic drug monitor ng purposes. Le but de cette étude était de développer et de valider une méthode analytique (HPLC­ MS/MS) pour déterminer les concentrations totales et libres de témocilline dans des échantillons biologiques . La méthode validée a été utilisée pour doser des échantillons sériques et tissulaires provenant de patients traités avec la témocilline.Méthodes: La précipitation des protéines sériques avec du méthanol et ultrafiltration ont été utilisés pour l'extraction de la fraction totale et libre de la témocilline, respectivement. L'extrait a été injecté dans un système LC-MS / MS pour la quantification de l 'antibiotique, en utilisant la ticarcilline comme standard interne.Résultats: Le dosage de la concentration de la fraction totale et libre de la témocilline était linéaire dans l'intervalle allant de 1 à 500 mg/L et de 0,5 à 300 mg/L, respectivement. L'exactitude et la précision inter et intra-jour étaient <13,9%. Les limites de quantification et de détection étaient de 1 et de 0,10 mg/L, et, de 0,5 et de 0,05 mg/L pour les concentrations des fractions totale et libre respectivement. Le rendement d'extraction de la témocilline était compris entre 85,80 et 99,40%. Aucun effet mémoire n'avait été observé et le phénomène de suppression d'ions était <36,2% dans les deux essais. La méthode a permis de doser la fraction libre et totale de témocilline dans des échantillons sériques d'un patient en hémodialyse , avec des résultats équivalents pour la fraction totale à ceux obtenus par une méthode HPLC-UV précédemment validée. Elle a ensuite été appliquée aux échantillons sériques et pancréatiques d'une patiente de soins intensifs chez qui nous avons observé une fraction libre élevée en relation avec une hypoalbuminémie et une concentration tissulaire supérieure à la concentration sérique.Conclusion: La méthode validée est spécifique, rapide, et sans interférences. Cette méthode peut être utilisée pour des études de pharmacocinétique pour suivi thérapeutique et pharmacologique de la témocilline.

Abstract

"A critical study of the demonic imagery that has been persistently embedded and codified in America's war culture. The authors examine "the devil myth" in both its past and present iterations and also highlight the counter-myth of the "trickster figure" whose democratic impulses have occasionally succeeded in countering the impulse towards demonization. To unveil the devil myth, the authors identify outward projections of evil onto the faces of America's enemies. They begin by scrutinizing the image of evildoers used to justify the global war on terror. It is difficult, they observe, to recognize this literalized image as a rhetorical construction subject to critical reflection without revisiting earlier manifestations of the devil myth in American history. Mythical projection is a cyclical process of political culture, they argue. Traces of earlier iterations of the devil myth carry into the present, but enemies are demonized anew in distinctive ways at each historical juncture of national crisis. To illustrate this process, the book includes chapters on demonized figures preceding the war on terror: witches, Indians, dictators, and reds. Each chapter shows how these emotionally loaded symbols have functioned as apparitions of dark foes that must be destroyed to redeem the nation's innocence. In this way, the book reveals how the subliminal figure of the devil haunts U.S. political culture so that war symbolically wards off evil in defense of, but at the cost of curtailing, its democratic soul. One of the study's underlying questions is how the nation can make peace with diversity instead of condemning it as a dark foe carrying the mark of evil. The book works toward an answer by discussing the creative and critical role of the democratic trickster"--

Abstract

Der hier neu in die Forschung eingeführte Begriff des ‚Cityscaping‘ charakterisiert den kreativen Prozess, in dem ein Stadtbild in verschiedenen Medien – Text, Film und Artefakten – erschaffen und repräsentiert wird. Er lenkt damit die Aufmerksamkeit von gebauten Stadträumen auf mentale Bilder von Städten. Im Zentrum steht einerseits die Frage, mit welchen literarischen, visuellen und akustischen Mitteln die räumliche Imagination der Rezipienten angeregt wird; andererseits wird untersucht, welche Semantik und Funktionen einem medial konstruierten Stadtbild beigelegt werden. An Beispielen antiker Texte und Kunstwerke, neuzeitlicher Literatur und Filme werden das künstlerische Potenzial und die Techniken der Semantisierung von Stadtbildern beleuchtet. Mit seinem interdisziplinären Ansatz macht der Band erstmals deutlich, wie stark die mentalen Bilder städtischer Räume in der Antike und Neuzeit von medialen Techniken geformt wurden. The term ‘cityscaping’ is here introduced to characterise the creative process through which the image of the city is created and represented in various media – text, film and artefacts. It thus turns attention away from built urban spaces and onto mental images of cities. One focus is on the question of which literary, visual and acoustic means prompt their recipients’ spatial imagination; another is to inquire into the semantics and functions that are ascribed to the image of a city as constructed in various media. The examples of ancient texts and works of art, and modern literature and films, are used to elucidate the artistic potential of images of the city and the techniques by which they are semanticised. With its interdisciplinary approach, the volume for the first time makes clear how strongly mental images of urban space, both ancient and modern, have been shaped by the techniques of their representation in media.
