Abstract

The current application of antibody-meditated targeted therapy against cancer has resulted in significant objective clinical responses, prolongation of survival and even cures. More than 20 mAbs have been approved for human use targeting a range of different cancers. However, a major drawback of mAb therapeutics is that a subset of patients does not initially respond and another initially responding subset develops resistance to further treatments. At the present time, there are no effective therapies for these subsets of cancer patients. The analyses of underlying mechanisms responsible for resistance are necessary to develop and generate new targeted therapies that overcome the resistance. Abstract

This book illustrates the intimate relationship between alveolar macrophages and Mycobacterium tuberculosis (M.tb.), and the former’s role in both innate and adaptive immunity against M.tb. It covers research done over the last decade. It also explores the role of macrophage death following infection with M.tb. in determining whether successful immunity is stimulated, or whether clinical disease develops; furthermore, the function of host lipid mediators in macrophage death modality are addressed. The book also illustrates how the balance between prostaglandins and lipoxins determines whether infected macrophages undergo apoptosis or necrosis, which is the ultimate factor in the outcome of infection. Finally, it is a synthesis of the authors’ recent studies and the studies of others to offer a new understanding of immunity to tuberculosis.

Abstract

The concept of delivering ‘magic bullets’ to treat diseases was first proposed by Paul Erlich in the early 1900’s.  The realization of this concept for the treatment of cancer occurred in the late 1990’s with the approval of monoclonal antibody therapies.  The use of monoclonal antibodies conjugated (linked) to potent cytotoxic agents (antibody-drug conjugates, ADCs) for specifically delivering cytotoxics to cancer cells was an obvious extension of antibody-based therapy.  ADCs have been under intense investigation for several decades; progress, however, has been limited due to toxicity or lack of improved efficacy compared to unconjugated cytotoxics.  More recently, linker technology and target selection have advanced such that several ADCs and immunotoxins are undergoing clinical testing or are approved for use.  This important volume gives the latest and most comprehensive information on the topic, describing different types of ADCs and immunotoxins for both hematologic and solid malignancies. Finally, the volume highlights the promise that this technology holds for diverse types of human cancer.
