Abstract

PCR methods for the detection of microbial pathogens have made relatively little impact in diagnostic microbiology laboratories due to the common decision to use expensive commercially produced tests rather than the cheaper alternative of developing one’s own tests or introducing tests developed by other workers. PCR Detection of Microbial Pathogens, Second Edition presents alternatives to commercially produced PCR methods to detect microbial pathogens. Although most of the chapters in this book are devoted to the detection of specific pathogens, the first  chapters in this book should appeal to anyone working in this field regardless of their particular interests. Although PCR tests can often be made to work with relatively little effort, it is often unclear how efficient the PCR test is, how inhibitory the specimen containing the pathogen of interest is and how the test can be quality controlled. All of which are of great importance in developing tests for diagnostic use. These topics are covered  in great depth at the beginning of the book. The main part of the book is devoted to describing methods for the detection of a wide range of pathogens and from widely different specimens and situations. Written in the highly successful Methods in Molecular Biology™ series format, chapters contain introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and notes on troubleshooting and avoiding known pitfalls.   Authoritative and accessible, PCR Detection of Microbial Pathogens, Second Edition serves microbiologists regardless of their particular interest because, when used together with the general principles, the sheer variety of procedures provided here enables the reader to design and introduce diagnostic tests in the laboratory with confidence.

Abstract

As imaging technologies and approaches have evolved, the scope of certain imaging techniques has moved far beyond the production of purely illustrative images or appealing time-lapse movies to providing the scientist with a rich range of ways to measure and quantify the biological process and outcome of gene expression.  In Imaging Gene Expression: Methods and Protocols, expert authors offer up-to-date approaches and protocols that scientists in the field have developed, which would benefit the broader scientific community. Divided in three convenient parts, this detailed book covers the output of a gene, namely the RNA molecules that are transcribed from the gene and the way by which these molecules can be tracked or quantified in fixed or living cells, protocols that focus on the gene, DNA, or chromatin, as well as a variety of ways by which nuclear processes intertwined with gene expression can be followed and quantified in living cells as well as approaches for studying several sub-nuclear structures found in eukaryotic cells.  Written in the highly successful Methods in Molecular Biology series format, chapters include introductions to their respective subjects, lists of materials and reagents, step-by-step, readily reproducible laboratory protocols, and tips on troubleshooting and avoiding known pitfalls.   Authoritative and up-to-date, Imaging Gene Expression: Methods and Protocols will serve researchers working toward imaging in the context of complete organisms.

Abstract

Extensive studies have been conducted on the identification, biogenesis, and processing of microRNA (miRNA) as well as research on the exact mechanism by which miRNAs bring about translational silencing of their targets. In addition, numerous publications point to an important role of miRNAs in development, reprogramming, epigenetics, pathogenesis of cancer, oncogenes and tumor suppressors, biomarkers of various disease onset, and regulation of adipogenesis and obesity; yet many questions still remain.  In MicroRNA Protocols, Second Edition, experts in the field provide up-to-date coverage of this diverse area of study.  The specific chapters of this edition are related to the analysis of miRNA, targets and expression profiling, various methods to determine its regulation of gene expression, the preparation and isolation of miRNAs in specific tissues, its detection in the saliva, and potential application in cosmetics, wound healing, and prostate cancer.  Written in the highly successful Methods in Molecular Biology™ series format, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and tips on troubleshooting and known pitfalls.   Fully updated and easy to use, MicroRNA Protocols, Second Edition aims to stimulate readers to explore diverse ways to understand the mechanisms in which miRNAs facilitate the molecular aspects of not only biomedical research but also a wide range of other research fields.

Abstract

Plants are amazing organisms to study, some are important sources for pharmaceuticals, and others can help to elucidate molecular mechanisms required for a plant’s development and its interactions with the biotic or abiotic environment. Functional genomics is vastly lagging behind the speed of genome sequencing as high-throughput gene function assays are difficult to design, specifically for non-model plants. Bioinformatics tools are useful for gene identification and annotation but are of limited value for predictions concerning gene functions as gene functions are uncovered best by experimental approaches. Virus-Induced-Gene-Silencing (VIGS) is an easy to use, fast, and reliable method to achieve down regulation of target gene expression. Virus-Induced Gene Silencing: Methods and Protocols provides detailed protocols for VIGS experiments in several plant species including model and non-model plants. Also included in this book are recently developed protocols for VIGS-derived microRNA production in the plant or protein over expression, as well as chapters devoted to summarizing the molecular mechanisms of VIGS action and the vector systems developed so far. Written in the successful Methods in Molecular Biology™ series format, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible protocols, and notes on troubleshooting and avoiding known pitfalls.   Authoritative and easily accessible, Virus-Induced Gene Silencing: Methods and Protocols serves as a valuable resource for researchers from diverse fields of plant biology interested in experimental approaches to analyzing gene functions.
