The hit-to-lead optimization process has evolved tremendously over the past decades, in order to achieve a better efficiency for pharma industry. This is indeed a key-step in the discovery of new drugs. Numerous techniques are now available to medicinal chemists to allow them to generate good quality hits and leads. High throughput screening is a frequent approach used in the identification of new hits, followed by a multi-disciplinary strategy that allows the parallel optimization of biological, physico chemical and pharmaco-kinetic properties. Following this approach, a new series of allosteric inhibitors of HCV NS5b polymerase was identified. The hit was subsequently optimized empirically, then rationally, once the active site has been characterized. The analogues synthesized during this work, together with the team effort, allowed to precise structure-activity relationships, white generating ADME data such as permeability and metabolic stability, which are key properties for the progress of the series. Le processus d'optimisation de molécules « hits » en « leads » a beaucoup évolué ces dernières années, afin de le rendre plus efficace pour l’industrie pharmaceutique. En effet, c'est une étape-clé dans la découverte de nouveaux médicaments. Maintenant de nombreuses techniques sont à la disposition des chimistes médicinaux afin de leur permettre de générer des hits et des leads de bonne qualité. Une approche fréquemment utilisée pour identifier un hit est le criblage à haut débit, suivi d'une approche multi-disciplinaire visant à optimiser de manière parallèle les propriétés biologiques, physico-chimiques et pharmacocinétiques. C'est cette approche qui a été suivie pour générer une nouvelle série d'inhibiteurs allostériques de la polymérase NS5b du virus de l'hépatite C. Le hit a ensuite été optimisé de manière empirique, puis plus rationnelle, une fois que le site actif de la cible a été caractérisé. Les analogues que nous avons synthétisés au cours de ce travail, associés à l'effort d'équipe du laboratoire, ont permis de préciser les relations structure-activités, tout en générant des données sur les paramètres ADME de perméabilité et stabilité métabolique, éléments déterminants pour l'avancement de la série.
