Abstract

The present thesis aimed at evaluating the potential of plasmid DNA vaccines directed against bovine herpesvirus 1 (BoHV-1). Five recombinant plasmids encoding for membranous or secreted forms of glycoproteins C and D as well as for the tegument protein VP8 of BoHV-1 were constructed and characterized in vitro and in vivo. A first evaluation of these constructs demonstrated the superiority of both plasmids encoding for the glycoprotein D (membranous and secreted forms). Those two constructs however showed a weak efficacy as compared to conventional vaccines that had been evaluated in a preliminary study. A second study showed the ability of DNA vaccines to successfully prime a BoHV-1 specific immune response. The immunity primed by DNA vaccines was considerably amplified by a single injection of a conventional inactivated marker vaccine adjuvanted with QuilA and aluminium hydroxide. The virological protection of the calves successively vaccinated with those two types of vaccines was similar to the one previously observed in calves vaccinated twice with the inactivated marker vaccine. A third experiment demonstrated the ability of a single intranasal administration of a recombinant adenovirus expressing BoHV-1 gD to boost the immune response primed by the DNA vaccines. The immune response induced by successive administrations of DNA vaccines and recombinant adenovirus was more equilibrated (Th1/Th2 ratio) than the immune response of the calves primed with plasmid DNA and boosted with the inactivated preparation. The boost with the recombinant adenovirus also contributed to the development of a higher mucosal immunity than the inactivated preparation. A fourth study showed that two slow-release systems, sustaining delivery of the DNA vaccines for about one month, allowed a better priming of the immune response than the discrete intradermal injections used so far. The system based on agarose implants allowed a priming of the BoHV-1 specific immune response with a si ngle handling of the calves. A fifth study finally concluded that the co-administration of oligodeoxynucleotides containing CpG motives enhanced both the precocity and the intensity of the immune response induced by DNA vaccines. Altogether, the observations conducted in the present thesis suggest that DNA vaccines will not be able to replace conventional vaccines but that they are complementary to them. The protocols combining DNA vaccines and inactivated marker vaccine or recombinant adenovirus open the prospect of an earlier vaccination of calves, even in the presence of a maternally derived immunity. The analysis of the virus amounts excreted after challenge and the values measured for immunological parameters across all the studies suggest the existence of significant correlation of the virological protection with neutralizing antibody titres and IgG2 titres.
