Abstract

Supercritical fluids behave either like a gas or a liquid, depending on the values of thermodynamic properties. This tuning of properties, and other advantageous properties of supercritical fluids led to innovative technologies. More than 100 plants of production size are now in operation worldwide in the areas of process and production technology, environmental applications, and particle engineering. New processes are under research and development in various fields. This book provides an overview of the research activities in the field of Supercritical Fluids in Germany. It is bas

Abstract

Among the most important and exciting current steps forward in geo-engineering is the development of coupled numerical models. They represent the basic physics of geo-engineering processes which can include the effects of heat, water, mechanics and chemistry. Such models provide an integrating focus for the wide range of geo-engineering disciplines. The articles within this volume were originally presented at the inaugural GeoProc conference held in Stockholm and contain a collection of unusually high quality information not available elsewhere in an edited and coherent form. Abstract

One of the most important objective in this text describes the strategies and approaches for the design of chemical processes. It covers economic (optimization) and environmental issues. The latest design strategies are described, most of which have been improved significantly with the advent of computers, mathematical programming methods, and artificial intelligence. Various methods are utilized to perform the extensive calculations and provide graphical results that are visualized easily, including the usage of computer programs for simulation and design optimization.

Abstract

Proteins continuously interact with each other to determine cell fate. Consequently, an examination of just when such protein-protein interactions occur and how they are controlled is essential for understanding the molecular mechanism of biological processes, elucidating the molecular basis of diseases, and identifying potential targets for therapeutic interventions. In Protein-Protein Interactions: Methods and Applications, leading experts describe in detail their highly successful biochemical, biophysical, genetic, and computational techniques for studying these interactions. Their readily reproducible methods demonstrate how to identify protein interaction partners, qualitatively or quantitatively measure protein-protein interactions, monitor protein-protein interactions as they occur in living cells, and determine interaction interfaces. The techniques described utilize a variety of cutting-edge technologies, including surface plasmon resonance (SRP), fluorescence resonance energy transfer (FRET), fluorescence polarization (FP), isothermal titration calorimetry (ITC), circular dichroism (CD), protein fragment complementation assays (PCA), various two-hybrid systems, and proteomics- and bioinformatics-based approaches, such as the Scansite program for computational analysis. Each time-tested protocol includes a background introduction outlining the principle behind the technique, lists of equipment and reagents, and tips on troubleshooting and avoiding known pitfalls. Authoritative and highly practical, Protein-Protein Interactions: Methods and Applications offers both beginning and experienced investigators a full range of the powerful tools needed for deciphering how proteins interact to form biological networks, as well as for unraveling protein-protein interactions in disease in the search for novel therapeutic targets.

Abstract

The study of RNA interference, editing, and modification has led to major shifts in our understanding of how genes are expressed and regulated. In RNA Interference, Editing, and Modification: Methods and Protocols, hands-on experimentalists describe in detail the protocols and assays they have developed to study these processes in most of the major biological systems in which they are known to occur-a wide range of organisms that includes worms, flies, trypanosomes, mammals, and plants. A historical overview of each subject is provided to put related chapters into a larger context. RNA interference (RNAi) has also emerged in recent years as an important and promising tool for gene discovery and reverse genetic engineering. Application of this cutting-edge technique is described for a range of experimental systems. Topics of interest include stable and transient RNA interference, RNA editing, gene silencing, bioinformatics, small noncoding RNAs, and RNomics. Special attention is given to methods for the identification and characterization of small RNAs, many of which are involved in RNA interference or modification. In addition, techniques used to study RNA editing mechanisms are elaborated for both systems that involve the conversion of one base to another and insertion-deletion editing. To promote the adaptation of their assays and approaches for use in other biological systems, the authors have included a range of methods representative of the major experimental systems in a given field. All protocols presented follow the successful Methods in Molecular Biology™ series format, each one offering step-by-step laboratory instructions, an introduction outlining the principle behind the technique, lists of equipment and reagents, and tips on troubleshooting and avoiding known pitfalls. State-of-the-art and highly practical, RNA Interference, Editing, and Modification: Methods and Protocols offers not only geneticists, molecular biologists, and bioinformaticists a comprehensive collection of readily reproducible techniques for the elucidation of gene function and the alteration of gene expression, but also describes how best they may be productively used to address outstanding mechanistic questions and adapted to novel biological systems.
