Hydroperoxide lyase is a cytochrome P450 that catalyses aldehydes sythesis from fatty acids hydroperoxides. In this work, the production yields of heterologous hydroperoxide lyase (expressed in Escherichia coli) have been optimized to reach approximately 15 mg of enzyme/liter. IMAC (Immobilized-metal affinity chromatography) purification techniques have been used thanks to the 6xHis tag genetically added to the enzyme. The study of reaction between hematin and hydroperoxides has demonstrated that this molecule has both homolytic and heterolytic hydroperoxide lyase activities.
