Abstract

Functional selectivity refers to the ability of different ligands acting at one receptor subtype to activate multiple signaling pathways in unique combinations; that is, one drug can be an agonist at pathway A and an antagonist or partial agonist at pathway B, and another drug can have the reverse profile. Functional selectivity has profound implications for drug development, for chemical biology, and for the design of experiments to characterize receptor function. In Functional Selectivity of G Protein-Coupled Receptors expert neuroscientists and pharmacologists review the work that demonstrated the existence of functional selectivity, placed it within a theoretical framework, and provided a mechanistic basis for the phenomenon. Abstract

Dopamine receptors are among the most validated drug targets for neuropsychiatric disorders such as schizophrenia and Parkinson’s disease, and are potential targets for other disorders such as substance abuse, depression, Tourette’s syndrome, and attention deficit hyperactivity disorder. The new knowledge brought from the creation and characterization of mice deficient in each of the subtypes and the development of increasingly subtype-selective agonists and antagonists has fueled many of the chapters new to this second edition, with the use of knock-out mice and subtype-selective drugs meriting discussion in stand-alone chapters. The field of G protein-coupled receptors has also advanced significantly since the first edition, with a model of GPCR signaling based on linear, compartmentalized pathways having been replaced by a more complex, richer model in which neurotransmitter effects are mediated by a signalplex composed of numerous signaling proteins, including multiple GPCRs, other types of receptors, such as ionotropic receptors, accessory and scaffolding proteins, and effectors. This second edition of Dopamine Receptors will be of interest to neurobiologists, pharmacologists and molecular biologists who study the brain, as well as neurologists working on psychiatric diseases.

Abstract

The ability to obtain health care is fundamental to the security, stability, and well-being of poor families. Government-sponsored programs provide temporary support, but as families leave welfare for work, they find themselves without access to coverage or care. The low-wage jobs that individuals in transition are typically able to secure provide few benefits yet often disqualify employees from receiving federal aid. Drawing upon statistical data and in-depth interviews with over five hundred families in Oregon, Karen Seccombe and Kim Hoffman assess the ways in which welfare reform affects the well-being of adults and children who leave the program for work. We hear of asthmatic children whose uninsured but working mothers cannot obtain the preventive medicines to keep them well, and stories of pregnant women receiving little or no prenatal care who end up in emergency rooms with life-threatening conditions. Representative of poor communities nationwide, the vivid stories recounted here illuminate the critical relationship between health insurance coverage and the ability to transition from welfare to work.
