Abstract

This volume provides a brief résumé of the history of testosterone research, from the early pioneers through to the most recent advances in the field. We discover how steroid hormones were first identified and how testosterone was shown to be essential for male development. Moving forward we explore how and where testosterone is produced, and how the body controls testosterone production. We then investigate the impact testosterone has on different body systems both during their development and function, and how perturbation of testosterone action is associated with disease. We complete our story with an exploration of the emerging roles of testosterone in clinical therapy, and the future potential for manipulation of the testosterone signaling system for human health benefit.

Abstract

This book serves as a tribute to the work of the many investigators who have attempted to understand the role of the androgen receptor (AR) in the development and progression of prostate cancer. The relationship between prostate cancer and androgen status was initially appreciated in the late 1800’s, and then awareness reawakened in the 1940’s by the studies of Charles Huggins and colleagues, for which the Nobel prize was awarded in 1966. The purpose of this book is to provide an up to date review of the molecular and cellular aspects of androgen action in prostate cancer. It includes an overview of the role of androgens in the development of the normal prostate and progression of prostate cancer. Our understanding of androgen metabolism and efforts to target androgen metabolism in the prevention and treatment of prostate cancer is presented. Androgen receptor structure/function relationships and the role of co-regulators are defined. Ligand-independent activation of the androgen receptor and the role of the androgen receptor during progression to the castration recurrent phenotype are discussed. Androgen-regulated genes are elucidated and their roles during prostate cancer progression are defined. Finally, the androgen receptor is presented as a viable target for novel therapies. Together, these reviews should give the reader a comprehensive conceptual framework of androgen action in prostate cancer.

Abstract

Combined oral contraceptives are the most convenient and accepted method of hormonal contraception. Nevertheless, the medical community and consumers constantly demand innovation, additional benefits during use and lower hormonal load despite the high safety profile of available products. At the Ernst Schering Research Foundation Workshop 52, new perspectives and mechanisms for tissue-selective, estrogen-free contraception were discussed. The aim of the workshop was to bring together experts in the field of molecular and pharmacodynamic action of progestins with clinicians and medical experts to discuss potential medical endpoints, physiological reactions and (bio)marker useful describing the tissue selectivity and the contraceptive action of new progestins in different target organs. A major factor for successful realization of these new concepts is a deeper understanding of local pharmacological responses to progestins in general and to new progestins in particular.

Abstract

Rapid actions of sex steroids play a critical role in mediating the biological outcome of target tissues. This book reviews the importance of these non-genomic effects in initiation and progression of breast and prostate cancers. Particular emphasis is placed on discussing how rapid steroid actions not only impact disease development and response to conventional therapies for these two types of cancers, but have also led to the identification of potential new therapeutic targets and treatment possibilities for cancer patients. The book reviews the fundamental basis of target development and the current status of these studies, which are providing valuable insights in key areas of breast and prostate cancers.
