Abstract

The book considers signaling events from the zygote embryo through to the blastocyst with relevant data from embryonic stem (ES) cells, including dialogue with the extracellular environment and with the maternal tract during the implantation process. Application of the knowledge described to improve the success of human and animal assisted conception is considered where appropriate, but the focus is largely on fundamental rather than applied cell/molecular biology, as this is the area that has historically been neglected. While the general features of metabolism during preimplantation development are well established, especially in terms of nutrient requirements, uptake and fate, remarkably little is known about early embryo signaling events, intracellular or intercellular, between individual embryos in vitro or with the female reproductive tract in vivo. This contrasts with the wealth of information on cell signaling in somatic cells and tissues, as a glance at any textbook of biochemistry illustrates. This lack of information is such that our understanding of the molecular cell biology of early embryos -- a prerequisite to defining the mechanisms which regulate development at this critical stage of the life cycle -- is seriously incomplete. This volume is the first to address this issue by describing the current state of knowledge on cell signaling during mammalian early embryo development and highlighting priority areas for research.

Abstract

T-type calcium channels are prevalent in every organ of the body, and are believed to play a part in a variety of physiological processes, including contraction, shape change, secretion, endo- and exocytosis, cell proliferation and differentiation, modulation of enzyme function, and cell cycle progression. As such, research on the roles of these channels in a wide range of diseases has attracted increasing attention in recent years. The purpose of this book is to present a series of unique and comprehensive reviews of our current understanding of T-type calcium channels at the level of the basic sciences as well as the clinical sciences. For key organ systems, information is provided on the correlation between the molecular, biophysical and pharmacological properties of the channels, their cellular mechanisms, and their potential roles in the pathogenesis of various diseases. Abstract

The book combines general concepts and methods to investigate calcium signalling in cells ranging from molecular biology approaches to manipulation of calcium in living cells. The focus within these methods in on the broad range of fluorescence imaging technology, in particular on optical sectioning techniques and fast image acquisition. In addition to these general guidelines there are application examples in a context beyond calcium signalling in two major fields: investigations of isolated cardiac myocytes and red blood cell related research. While the cellular cardiology section provides snapshots of certain calcium signalling aspects, the red blood cell part presents an overview from the functional identification of calcium-channels to a concept of physiological and pathophysiological relevance.

Abstract

Almost 25 years ago, the first mammalian transient receptor potential (TRP) channel was cloned and published. TRP channels now represent an extended family of 28 members fulfilling multiple roles in the living organism. Identified functions include control of body temperature, transmitter release, mineral homeostasis, chemical sensing, and survival mechanisms in a challenging environment. The TRP channel superfamily covers six families: TRPC with C for “canonical”, TRPA with A for “ankyrin”, TRPM with M for “melastatin”, TRPML with ML for “mucolipidin”, TRPP with P for “polycystin”, and TRPV with V for “vanilloid”. Over the last few years, new findings on TRP channels have confirmed their exceptional function as cellular sensors and effectors. This Special Book features a collection of 8 reviews and 7 original articles published in “Cells” summarizing the current state-of-the-art on TRP channel research, with a main focus on TRP channel activation, their physiological and pathophysiological function, and their roles as pharmacological targets for future therapeutic options.

Abstract

Alzheimer’s disease (AD) is an age-related neurological disease that affects tens of millions of people, in addition to their carers. Hallmark features of AD include plaques composed of amyloid beta, as well as neurofibrillary tangles of tau protein. However, despite more than a century of study, the cause of Alzheimer’s disease remains unresolved. The roles of amyloid beta and tau are being questioned and other causes of AD are now under consideration. The contributions of researchers, model organisms, and various hypotheses will be examined in this Special Issue.
