Only recently have we come to ask what role the male might play in pregnancy loss and embryo implantation failure, beyond contributing an abnormal set of paternal chromosomes at fertilization. This volume explores the recent evidence that these conditions might also be related to the transmission of previously unrecognized chromosomal micro-deletions and duplications, or via the epigenetic dysregulation of early embryonic gene function by spermatozoal microRNAs or alterations in sperm DNA methylation patterning, or by DNA damage secondary to either reactive oxygen species or environmental toxicants. In addition, the composition of seminal plasma has been found to be highly complex, containing many factors that play important roles in altering the uterine environment and the female immune system permissive of embryo implantation and trophectoderm outgrowth leading to successful pregnancy. Abstract

The book considers signaling events from the zygote embryo through to the blastocyst with relevant data from embryonic stem (ES) cells, including dialogue with the extracellular environment and with the maternal tract during the implantation process. Application of the knowledge described to improve the success of human and animal assisted conception is considered where appropriate, but the focus is largely on fundamental rather than applied cell/molecular biology, as this is the area that has historically been neglected. While the general features of metabolism during preimplantation development are well established, especially in terms of nutrient requirements, uptake and fate, remarkably little is known about early embryo signaling events, intracellular or intercellular, between individual embryos in vitro or with the female reproductive tract in vivo. This contrasts with the wealth of information on cell signaling in somatic cells and tissues, as a glance at any textbook of biochemistry illustrates. This lack of information is such that our understanding of the molecular cell biology of early embryos -- a prerequisite to defining the mechanisms which regulate development at this critical stage of the life cycle -- is seriously incomplete. This volume is the first to address this issue by describing the current state of knowledge on cell signaling during mammalian early embryo development and highlighting priority areas for research.

Solid tumors frequently contain areas of oxygen deprivation (hypoxia) due to rapid cell proliferation and/or vascular insufficiency. The presence of hypoxic domains typically correlates with poor patient prognosis, due to the relative resistance of hypoxic cells to conventional cancer therapies and effects of O2 availability on disease progression. The response of malignant cells to hypoxia has been the focus of intense research over the last decade. In this issue of Current Topics in Microbiology and Immunology, the authors present articles describing the impact of hypoxia on components of the tumor microenvironment (such as endothelial cells, inflammatory cells, and tumor associated fibroblasts), the expression of unique microRNAs, tumor cell differentiation status, and metastasis.
