Endoplasmic reticulum (ER) is an intracellular organelle responsible for protein folding and assembly, lipid and sterol biosynthesis, and calcium storage. A number of biochemical, physiological, or pathological stimuli can interrupt protein folding process, causing accumulation of unfolded or misfolded proteins in the ER lumen, a condition called “ER stress”. To cope with accumulation of unfolded or misfolded proteins, the ER has evolved a group of signaling pathways termed “Unfolded Protein Response (UPR)” or “ER stress response” to align cellular physiology. To maintain ER homeostasis, transcriptional regulation mediated through multiple UPR branches is orchestrated to increase ER folding capacity, reduce ER workload, and promote degradation of misfolded proteins. In recent years, accumulating evidence suggests that ER stress-triggered transcriptional reprogramming exists in many pathophysiological processes and plays fundamental roles in the initiation and progression of a variety of diseases, such as metabolic disease, cardiovascular disease, neurodegenerative disease, and cancer. Understanding effects and mechanisms of ER stress-associated transcriptional reprogramming has high impact on many areas of molecular genetics and will be particularly informative to the development of pharmacologic avenues towards the prevention and treatment of modern common human diseases by targeting the UPR signaling. For these reasons, ER stress response and transcriptional reprogramming are a timely and necessary topic of discussion for Frontiers in Genetics.

This book collects some recent scientific data on the role of polyphenols in various pathological contexts, ranging from viral infection to metabolic or chronic disorders such as diabetes and renal fibrosis. The included studies showed both in vitro and in vivo evidence concerning positive effects of polyphenols in inflammation, apoptosis, and oxidative stress. The investigated molecules include: verbascoside from Olea europea, curcumin from Curcuma longa, phenolic acids from Antirhea borbonica, and phlorotannins from marine algae. Finally, the main flavonoids present in the human diet and their anti-inflammatory and anti-cancer roles were also discussed. The scientific data confirm the importance of the plant kingdom, both marine and terrestrial, in the search for new compounds with potential benefits for humans.

Cancer has been a patient-specific and difficult-to-treat disease for decades, resulting in more deaths since 1900 than all other diseases except cardiovascular diseases. As societies around the world continue to shift towards an aging population, the social and economic burden created by cancer will only rise in the coming decades, necessitating continued improvement in our cancer therapies. Remarkably, in the late 1800s, bone surgeon William Coley serendipitously discovered that bacteria could be administered to patients as an effective (and sometimes toxic) form of cancer therapy known as "Coley's Toxins". His discoveries unknowingly led to two fields of cancer therapy that have been in development for decades and are now leading to significant improvements in therapy for cancer patients: immune-based and toxin-based therapies for cancer. Articles included here discuss the discoveries that emerged from Coley's Toxins that enable us to harness the immune system and microbial toxins to combat cancers, as oncology shifts from a field dominated by chemotherapy for most of the 20th century to biologic therapies that will dominate the 21st century.
