Abstract

Plants synthesize a wide variety of unique glycan structures which play essential roles during the life cycle of the plant. Being omnipresent throughout the plant kingdom, ranging from simple green algae to modern flowering plants, glycans contribute to many diverse processes. Glycans can function as structural components in the plant cell wall, assist in the folding of nascent proteins, act as signaling molecules in plant defense responses or (ER) stress pathways, or serve within the energy metabolism of a plant. In most cases, glycans are attached to other macromolecules to form so-called glycoconjugates (e.g. glycoproteins, proteoglycans and glycolipids), but they can also be present as free entities residing in the plant cell. Next to the broad, complex set of glycans, plants also evolved an elaborate collection of lectins or proteins with a lectin-like domain, which can recognize and bind to endogenous (plants-own) or exogenous (foreign) glycans. Though still poorly understood in plants, the dynamic interactions between lectins and carbohydrate structures are suggested to be involved in gene transcription, protein folding, protein transport, cell adhesion, signaling as well as defense responses. As such, a complex and largely undetermined glycan-interactome is established inside plant cells, between cells and their surrounding matrix, inside the extracellular matrix, and even between organisms. Studying the biological roles of plant glycans will enable to better understand plant development and physiology in order to fully exploit plants for food, feed and production of pharmaceutical proteins. In this Research Topic, we want to provide a platform for articles describing the latest research, perspectives and methodologies related to the fascinating world of plant glycobiology, with a focus on following subjects: 1. Identification and characterization of plant glycans, their biosynthetic and degradation enzymes 2. Characterization of plant lectins and glycoproteins 3. Abstract

The first chapter of the book reports on the management of Langerhans cell histiocytosis (LCH)-induced central diabetes insipidus and its associated endocrinological/neurological sequelae in the national survey. The next chapter addresses DI and head injuries. Next, the management of neuroendocrine instability during maintenance of potential organ donors is described. Organ transplants have gradually increased worldwide. To have maintenance of appropriate potential organs, AVP is needed. Furthermore, nephrogenic DI-the potential therapeutic drugs and analysis of membrane protein stability is the topic of the next two chapters, followed by new insights into the diagnosis and management of pregnancy-related DI. The seventh chapter reports on the problems with differential diagnosis in a case of central DI in a female patient with bipolar disorder. The lithium treatment usually resulted in nephrogenic DI. Finally, over the last years, the development of MRI imaging on the pituitary gland with the stalk and hypothalamus has advanced. The final chapter interprets imaging techniques in DI in detail.

Abstract

The fundamental treatment and management goals in diabetes mellitus are to control and normalize blood glucose levels and to prevent diabetic complications. It also includes maintaining normal growth and development and normal body weight. Proper diet, regular exercise, weight control and different therapeutic agents are the mainstays of diabetic care and management. Weight reduction and exercise have been shown to improve tissue sensitivity to insulin and allow its proper use by target tissues. It is obvious that medical management and goals of therapy for diabetes mellitus have changed since the publication of the Diabetes Control and Complications Trial in 1993. Recent studies have shown that the risk of developing retinopathy can decrease by 76% in properly managed diabetic patients when compared to control group and that clinical and laboratory signs and symptoms of nephropathy and neuropathy can also decrease by 54 to 60%. Modern approaches to the management of diabetes mellitus embrace holistic options and this book addressed various approaches in the management of diabetes mellitus.

Type 2 diabetes is now a global serious health problem. Patients with type 2 diabetes have 2-4 times higher risk of cardiovascular and renal complications, morbidity and mortality. Abstract

Since variolation, conventional approaches to vaccine development are based on live-attenuated, inactivated or purified pathogen-derived components. However, effective vaccines against global health threats such as HIV, parasite infections and tumors are difficult to achieve. On the other hand, synthetic vaccines based on immunogenic epitopes offer advantages over traditional vaccines since they are chemically defined antigens free from deleterious effects. Additionally, in contrast to live-attenuated vaccines, they do not revert to virulence in immunocompromised subjects, and different from genetic vaccines, they do not involve ethical questions. Traditional vaccines contain PAMPs and induce strong immune responses, while recombinant vaccines are less potent. In spite of the immunogenic weakness previously attributed to epitope-based vaccines a synthetic vaccine containing a 17 amino acid-epitope of the Pseudomonas aeruginosa Type IV pilus exceeded the protective potential of its cognate protein composed of 115 amino acids. Therefore, the efficacy yield of a synthetic vaccine can be potentiated by using the proper combination of target epitopes. Recent advances in adjuvant development, immunogen platforms for DNA vaccines and viral vectors also contributed to optimize immunogenicity. Another constraint to the use of epitope vaccines was their restriction to some MHC or HLA phenotypes. However, epitopes containing 20 or less amino acids of Plasmodium falciparum and Leishmania donovani bind to multiple HLA-DR and MHC receptors. Thus synthetic epitope vaccines may better meet the requirements of the regulatory agencies since they have lower costs and are easier to produce. The classical experimental approach for the development of an epitope-based vaccine involves the use of recombinant domains or overlapping 15-mer peptides spanning the full length of the target antigen, and the analysis of the induced antibody and/or T cell immune responses in vitro or in vivo. On the other hand, in silico tools can select peptides that are more likely to contain epitopes, reducing the number of sequence candidates. T cell epitope prediction dates back to 1980s, when the first algorithm was developed based on the identification of amphipathic helical regions on protein antigens. Since then, new methods based on MHC peptide-binding motifs or MHC-binding properties have been developed. The recent reverse vaccinology concept uses high-throughput genome sequencing and bioinformatics tools to identify potential targets of immune responses. The feasibility of this approach was shown for the first time in the design of a vaccine against Neisseria meningitides that is now in phase III clinical trials. In addition, different computational tools allow the determination of crucial gene(s) through comparative analyses between different pathogenic strains Alternatively, carbohydrates have been considered as key targets in developing safe and effective vaccines to combat cancer, bacterial and viral infections. Tumor associated carbohydrate antigens can be coupled covalently to protein carriers to target MHC receptors and improve immunogenicity and have reached already pre-clinical and clinical studies. In light of the recent availability of genomic tools, we believe that in the near future an increasing number of vaccine candidates, composed of defined epitopes, will be available for synthetic vaccines showing improved protection.

Abstract

The cell surface of fungi, bacteria and sea organisms is highly glycosylated. These glycans are oligo- or polysaccharide molecules that can be secreted or attached to protein or lipids forming glycoconjugates. They present extraordinary structural diversity that could explain their involvement in many fundamental cellular processes, including growth, differentiation and morphogenesis. Considerable advances have been made on the structural elucidation of these glycans. Their primary structures were determined based on a combination of mass spectrometry and NMR spectroscopy techniques. The combination of these sensitive and powerful techniques has allowed us to increase our structural knowledge of a wide variety of glycans expressed by different fungi, bacteria and sea organisms.
