Abstract

This book provides a 2012 reference for state-of-the-art gene transfer technology and the different aspects of its clinical translation with a focus on European-based initiatives. As examples of successful outcomes, recent clinical trials are presented together with Ethical, Safety and Regulatory issues, which are discussed. The broad range of various technologies is covered whether addressing direct in vivo gene transfer like with AAV, Adeno or non-viral vectors or ex-vivo genetically engineered cells including induced pluripotent stem cells (iPS) with integrating vectors such as retrovirus, lentivirus or transposon-derived systems. The critical path to clinical implementation is covered in the second part describing currently available tools - such as molecular imaging, ex-vivo organ cultures and high-throughput technologies used for evaluation of criteria towards a go-or-no go decision to move to the clinic; in addition, utmost salient biosafety and immunotoxicology aspects are discussed. This book is ideal for postgraduates, undergraduates, scientists, clinicians, regulators and patients' advocacy groups looking for states-of-the-art information as well as emerging prospects - including gene targeting and homologous recombination - in gene transfer intended for clinical translation.

Abstract

Whether to assess the function of new genes identified from the Human Genome Project or to apply gene therapy successfully, it is often necessary to deliver genes to specific cells. In Gene Delivery to Mammalian Cells, highly experienced researchers describe in great detail methods that have proven most useful in delivering genes to mammalian cells. Volume 1: Nonviral Gene Transfer Techniques focuses on gene delivery by a variety of chemical and physical methods, including ultrasound, biolistics, peptides, PNA clamps, liposomes, microinjection, electroporation, particle bombardment, and hydrodynamics. Each protocol contains step-by-step instructions, along with background notes, equipment and reagent lists, and tips on troubleshooting and avoiding known pitfalls. Introductory chapters review the delivery methods presented, discussing their advantages and disadvantages, how they have been used successfully for gene delivery, and the future of their technology. An accompanying volume, Volume 2: Viral Gene Transfer Techniques, details procedures for delivering genes to cells in vitro and in vivo. Comprehensive and cutting-edge, Gene Delivery to Mammalian Cells, volumes 1 and 2, offers biomedical specialists in industry and academia an unrivaled collection of reliable methods for the chemical, physical, and viral delivery of genes to mammalian cells, all prepared to save laboratory time and ensure experimental success.

Abstract

In Gene Therapy Protocols, Volumes 1 & 2, internationally recognized investigators describe cutting-edge laboratory techniques for the study of Production and In Vivo Applications of Gene Transfer Vectors (Volume 1) and Design and Characterization of Gene Transfer Vectors (Volume 2). The field of gene therapy has undergone remarkable advances, promising to impact human healthcare significantly in the twenty-first century. Today’s technologies can deliver genetic material safely and effectively to cells to slow or halt the progression of disease, and to help repair or regenerate damaged or lost tissues. In this first volume of Gene Therapy Protocols: Production and In Vivo Applications of Gene Transfer Vectors, readers will find a comprehensive resource of current and emerging methods for the production of viral and non-viral gene transfer vectors, as well as detailed protocols for critical applications in stem cell biology, cancer, diabetes, HIV and tissue engineering. This first volume of the new and completely revised third edition of Gene Therapy Protocols will prove a necessary tool for graduate students and postdoctoral fellows and invaluable to basic and clinical researchers in both industry and academia.

Abstract

This book focuses on recent trends of research on alginate-based biomaterials in Drug Delivery strategies and biomedical engineering. It contains the widely used alginate-based biomaterials as micro to nano-controlled drug delivery (oral, ocular delivery, topical delivery, etc.) and its fabrication technology, characterization, and biomedical aspects (such as cancer therapy, tissue engineering, gene delivery, vaccine delivery, enzyme immobilization, wound healing, dental applications, etc.) in a single book. The chapters cover updated information, current research trends, informatics, and all aspects of applications. Alginate is a U.S. Food and Drug Administration (FDA)-approved natural biomaterial and has diverse biomedical applications. In recent years, researchers and scientists are working on the alginate-based drug delivery systems that have been designed and characterized as a matrix, micro to nanocarriers, fibers, composite/scaffolds, etc. Alginate has versatile properties such as biodegradable, biocompatible, nontoxic, and easily available. This book especially highlights both the drug delivery strategies and biomedical engineering aspects such as controlled drug delivery, drug targeting to the site of action, cancer therapy, gene and vaccine delivery, enzyme immobilization, tissue engineering, and regenerative medicine. .
