The goal of this work was to identify risk factors for bladder cancer. The main risk factors are smoking and occupational exposure to aromatic amines, nitrosamines polycyclic aromatic hydrocarbons and arsenic. We must also consider infect ions such as Schistosoma haematobium, HPV, or HIV. In addition to these exogenous factors, genetics play an important role in the genesis of bladder cancer. Polymorphisms in the signaling pathway Sonic hedgehog. within the P2R Y5 gene or hOGG1 , Within metabolic enzymes such as NAT1, N AT2 GST were identified as polymorphisms which may generate bladder cancer. Moreover people with a slow acetylation arc more at risk of cancer because the toxins remain n longer in contact with the bladder.Regarding the exogenous risks, it is important to educate people at risk" to try to prevent cases of cancer. On the other side, regarding polymorphisms. it is essential to take steps to detect and closely monitor these cases. Pour répondre à la problématique de ce mémoire qui était de relever les facteurs de risque du cancer de la vessie, il a été intéressant de développer les principaux facteurs, à savoir le tabagisme et l 'exposition professionnelle aux amines aromatiques, nitrosamines, hydrocarbures aromatiques polycycliques et arsenic. Il faut également prendre en compte les éventuelles infections telles que les Schistosomes, papillomavirus ou HIV.Outre ces événements exogènes, la génétique joue un rôle important dans la genèse du cancer de la vessie. Les polymorphismes au sein de la voie de signalisation Sonic Edgehog, au sein du gène P2RY5 ou HOGG1 , au sein d'enzymes de métabolisation telles que NAT1 , NAT2, GST ont été relevés comme étant des polymorphismes susceptibles de générer un cancer de la vessie. De plus, les acétylateurs lents seraient plus exposés au risque de cancer que les acétylateurs rapides car les toxines restent plus longtemps en contact avec la vessie.En ce qui concerne les risques exogènes, il est important de sensibiliser les « personnes à risque » pour tenter de prévenir des cas de cancer. Par contre, en ce qui concerne les polymorphismes, il est essentiel de prendre les mesures nécessaires afin de les dépister et de surveiller étroitement ces cas.
